摘要
为研究内毒素(ET)致大鼠肝细胞凋亡变化、对肝脏Caspase-3蛋白表达的影响及阳离子A(CA)的保护效应。将试验动物随机分为3组:对照组(Ⅰ组)、ET组(Ⅱ组)和CA保护组(Ⅲ组)。3组动物经相应处理后分别在3、4、8、12 h采集肝脏检测肝细胞凋亡情况和Caspase-3蛋白的表达情况。结果表明,肝细胞凋亡与Caspase-3蛋白的表达成正相关的关系。ET可上调Caspase-3蛋白在肝脏的表达,诱导肝细胞凋亡,最终导致肝脏受损;而CA则能明显下调Caspase-3在肝脏的表达,抑制肝细胞凋亡,对由ET诱导的肝损伤具有显著的保护效应。
The effect of endotoxin on expressions of Caspase-3 and the protective effect of Cation A in the liver were studied in this paper.Seventy-two Wistar rats were randomly divided into three groups: the normal controls group(groupⅠ),ET treatment group(groupⅡ) and Cation A(group Ⅲ).The CA was used to study the protective effects of the ET on apoptosis.At three,four,eight and twelve hours after the animals were treated,hepatocyte apoptosis and the expression of Caspase-3 in liver were detected.The results showed that it was the positive correlation relationship between the hepatocyte apoptosis of hepatic cell and expression of Caspase 3.The ET could up-regulate the expression of Caspase-3 in liver,then induced the hepatocyte apoptosis,eventually leaded to liver damage.Whereas CA could significantly down-regulate the Caspase-3 expression and inhibited the hepatic cell apoptosis,also had significant protective effect on liver injuries caused by ET.
