摘要
以培养的人脐静脉内皮细胞(EC)为损伤模型,观察到氧化修饰的低密度脂蛋白(O—LDL,100μg/ml和叔丁基脂氢过氧化物(tbooH,10^(-2)mM)对内皮细胞的硒—谷胱甘肽过氧化物酶(SeGSH—Px)的活性,前列腺环素(PGI_2)的生成,脂质过氧化物(Lpo)含量及SeGSH—Px/Lpo比值有明显的时间依赖效应,即随作用时间的延长,O—LDL和tbooH使EC的SeGSH—Px活性下降,PGI_2生成减少,SeGSH—Px/Lpo比值降低;而Lpo含量则增加。提示O—LDL对EC的损伤实质是脂质过氧化物的作用。
Oxidized low density lipoprotein may induce damages to endothelial cells. There is a close relationship between the endothelial cell injury and the development and progressing of atherosclerosis. Here we compared the effects of oxidatively modified low density lipoprotein ( O-LDL ) and tert-butyl hydroperoxide ( tbooH ) on the production of prostacyclin ( PG I2 ) , activity of selenium-glutathione per-oxidase ( SeGSH-Px ) , content of lipoperoxide ( Lpo ) and ratio of SeGSH-Px/Lpo in cultured human umbilical cord vein endothelial cells ( EC ) at various intervals of incubation time (Oh, 24h, 48h, 72h ) .Results of the present study showed thatO-LDL and tbooH both induced time-dependent alterations in the PGl2 prodution, SeGSH-Px activity、 Lpo content and SeGSH-Px/Lpo ratio in EC. The effects of tbooH on EC were consistent with those of O-LDL. The results obtained indicate that the effects of O-LDL on EC are similar to those of tbooH, which suggests that the injury of EC by O-LDL is in fact the result of effects of Lpo.
