摘要
目的:探讨五没食子酰葡萄糖(PGG)诱导鼻咽癌细胞的凋亡作用。方法:PGG孵育人鼻咽癌5-8F细胞株48h后,采用倒置相差显微镜观察细胞形态;PGG孵育人鼻咽癌5-8F细胞株24h后,采用流式细胞仪分析细胞早期凋亡。结果:25,50和75μmol/L PGG孵育5-8F细胞48h后,贴壁活细胞减少,漂浮死亡细胞增多。PGG孵育24h后,细胞早期凋亡率分别为(4.00±0.56)%、(6.97±0.25)%和(10.23±0.71)%,与对照组(1.53±0.15)%比较,差异有统计学意义(P<0.05)。结论:PGG具有诱导鼻咽癌细胞凋亡的生物学作用。
Objective : To investigate the effect of 1,2,3,4,6-penta-O-galloy-D-glucose(PGG) on apoptosis of nasopharyngeal carcinoma cells. Methods: PGG was incubated with human nasopharyngeal carcinoma 5-8F cell line for 48 h, cell morphology was observed by inverted phase contrast microscope. PGG was incubated with human nasopharyngeal carcinoma 5-8F cell line for 24 h, the early apoptotic cells were analyzed by Flow Cytometry. Results. 25, 50 and 75 μmol/L PGG were respectively incubated with 5-8F cell line for 48 h, the adherent living cells reduced, floating dead cells increased. Incubated for 24 h, early apoptosis cells were (4 ± 0.56)%, (6.97 ± 0.25)% and (10.23± 0.71)% in 25, 50 and 75 μmol/L PGG groups, compared with the control group (1.53 ± 0.15)%, the difference was statistically significant (P〈0.05). Conclusion. PGG has biological effects of inducing apoptosis of nasopharyngeal carcinoma ceils.
