电针对阿尔茨海默病大鼠mTOR信号通路相关蛋白表达的影响 被引量:3

Effect of Electroacupuncture on Expression of mTOR Signal Pathway Associated Protein in Alzheimer's Disease Rat

摘要 目的探讨电针对阿尔茨海默病(AD)大鼠海马组织mTOR信号通路相关蛋白表达的影响。方法将大鼠随机分为正常组、假手术组、模型组和电针组,采用Morris水迷宫检测空间学习和记忆能力的变化。HE染色观察各组海马CA1区组织形态变化;Western blot检测p-mTOR、p-p70S60K和p-4E-BP1的水平。结果电针治疗可明显改善AD大鼠学习和记忆力,减轻海马CA1区组织形态的变化,并可降低海马组织p-mTOR和p-p70S60K的表达,而对p-4E-BP1的表达无明显影响。结论电针治疗可能通过调控mTOR信号通路及下游蛋白p70S6K的表达,对抗Aβ诱导的神经元损伤,提高AD大鼠学习记忆功能。 Objective To explore the effect of electroacupuncture on the expression of mTOR signaling pathway related protein in hippoeampal tissue of Alzheimer's disease (AD) rats. Methods The rats were randomized into the normal group,the sham operation group, the model group, and the electroacupuncture treatment group. The learning and memory abilities were detected by Morris water maze. Morphologic changes of hippocampal CA1 region were observed by HE staining. Level of p mTOR,p- p70S60K,and p 4E- BP1 were determined by Western blot analysis. Results Electroaeupuncture treatment could significantly improve the learning and memory function of AD rats, reduce the morphology changes of hippocampal CA1 area,and reduce the expression of p mTOR and p - p70S60K in hippocampus. It had no effect on the expression of p - 4E BP1. Conclusion Electroacupuncture could antagonize against β - amyloid protein induced neuronal damage, and improve the learning and memory function of AD rats through regulating the expression of mTOR signaling pathway and its downstream protein pTOS6K.
