摘要
以悬浮CHO DG44细胞为对象,采用central composite响应面分析法,以活细胞密度为评价指标,确定了3种对细胞生长有明显促进作用的培养基关键组分——胰岛素、转铁蛋白及亚硒酸钠(简称ITS)的最优浓度及其相互影响模式,并设计了一种有效的低成本ITS替代混合物。同时在添加25 mg/L硫酸葡聚糖条件下成功解决细胞高密度悬浮培养的成团问题,为CHO细胞低成本无血清培养基的研制提供了理论依据。
With suspension Chinese Hamster Ovary cell lines(CHO DG44) as the object of study,a serum free medium for the cultivation of CHO cells in suspension was formulated by using central composite response surface methodology.As Insulin,transferrin,and sodium selenite(ITS) were identified as the most significant factors,the response surface methodology was used to determine the optimal levels of these factors,where the evaluating index was viable cell density.And an inexpensive substitution of ITS for CHO cells suspension culture was formulated.Also the clumping of cells was resolved by adding 25 mg/L dextran sulfate to medium,which provided a basis for inexpensive serum free medium for CHO cells.
