摘要
采用酶法辅助超声提取高寒香菊花多糖。通过单因素试验及正交试验分别对酸性蛋白酶,纤维素酶超声提取高寒香菊花花多糖的最优工艺条件进行了研究。结果表明:酸性蛋白酶超声辅助提取高寒香菊花多糖最佳工艺条件为:酶解温度为45℃,酶用量0.5%,pH 4.0,酶解时间2.5 h,高寒香菊花多糖提取率为8.25%,纯度为23.30%;纤维素酶超声辅助提取高寒香菊花多糖最佳工艺条件为:酶解温度为45℃,酶用量2.0%,pH 5.0,酶解时间1.0 h,高寒香菊花多糖提取率为7.84%,纯度为13.25%。
Coreopsis basalis flower polysaccharide was obtained by enzyme-assisted ultrasonic extraction. The optimal extraction process conditions of polysaccharides from Coreopsis basalis flowers were researched with acid protease and cellulose respectively by single factor experiment and orthogonal experiment. The results showed that the optimal extraction process conditions of acid protease was as follows; enzymolysis temperature 45 ℃, enzyme 0.5%, pH 4.0, enzymolysis time 2.5 h. Under these conditions, the yield of Coreopsis basalis flower polysaccharide was up to 8.25%, purity was 23.30%. The optimal extraction process conditions of cellulose was as follows; enzymolysis temperature 45 ‰, enzyme 2.0%, pH 5.0, enzymolysis time 1.0 h. Under these conditions, the yield of Coreopsis basalis flower polysaccharide was up to 7.84%, purity was 13.25%.
