红景天中苷及其醇类化合物抗辐射有效活性成分体外筛选研究 被引量:9

摘要 目的比较研究红景天中6种苷及醇类化合物红景天苷、异槲皮苷、酪醇、洛塞琳、熊果苷、络塞维的体外抗辐射活性。方法给予人淋巴细胞10 Gy60Coγ射线一次性照射,构建辐射防护模型。以MTS法检测6种苷及醇类化合物对辐射损伤人淋巴细胞增殖的保护作用。结果与正常组比较,4、6、8、10 Gy60Coγ模型组人淋巴细胞存活率显著降低,其中10Gy60Coγ模型组存活率为(55.43±4.69)%。与模型组比较,异槲皮苷(12.5μg.ml-1)、红景天苷(12.5μg.ml-1)、络塞维(12.5μg.ml-1)、洛塞琳(25μg.ml-1)、熊果苷(12.5μg.ml-1)、酪醇(50μg.ml-1)给药组均能够显著提高辐射损伤人淋巴细胞的增殖活性。结论红景天中苷及醇类成分对60Coγ射线照射所致人淋巴细胞损伤具有一定的保护作用,其中浓度为25μg.ml-1的络塞维抗辐射作用最强。 Objective To make a comparative study of the radioprotective activity of six glycosides or alco- hols from Rhodiola in vitro , including salidroside, lsoquercitrin, tyrosol, rosarin, Arbutin and rosavin. Methods Lym- phocytes (AHH-1) were exposed to 10 Gy 60Coγ ray to induce a radiation injury model before MTS assay was used to detect the proliferation activity of AHH-1 after being treated by six kinds of glycosides or alcohols from Rhodiola. Results Compared with the normal group,4,6,8 and 10 Gy model groups showed obvious low survivability of AHH- 1. The survival rate of 10 Gy model group was ( 55.43 ± 4.69 ) %. Compared with the model group, isoquercitrin ( 12.5 μg ·ml^-1 ) ,salidroside( 12.5 μg ·ml^-1 ) ,rosavin( 12.5 μg ·ml^-1 ), rosarin (25 μg ·ml^-1 ), Arbutin ( 12.5 μg ·ml^-1) and tyrosol (50 μg ·ml^-1) could obviously improve the viability of AHH-lafter radiation. Conclusion Glycosides or alcohols from Rhodiola had anti-radiation activities on AHH-1, and at the concentration of 25 μg ·ml^-1 ,rosavin had the best anti-radiation effect among the six components.
