Simultaneous Determination of Chlorogenic Acid Geniposide and Baicalin in Qingre Jiedu Soft Capsule by HPLC

目的建立HPLC法同时测定清热解毒软胶囊中绿原酸、栀子苷和黄芩苷的含量。方法 Agilent EclipseXDB-C18色谱柱(4.6 mm×150 mm,5μm),流动相为甲醇(A)-2%冰醋酸溶液(B),梯度洗脱;流速1.0 ml.min-1,检测波长254 nm,柱温30℃。结果绿原酸、栀子苷和黄芩苷的进样量与峰面积分别在0.010 92～0.4368μg(r=0.9999),0.010 20～0.4080μg(r=0.9999)和0.042 56～1.7024μg(r=0.9999)呈良好的线性关系;平均加样回收率分别为99.97%,99.02%,98.70%,RSD为1.57%,1.42%,1.52%。结论本方法操作快速、结果准确,专属性强,可用于清热解毒软胶囊的质量评价。

Objective To establish an HPLC method for determination of chlorogenic acid, geniposide and baicalin in Qingre Jiedu soft capsules. Methods Agilent Eclipse XDB-Cls was used with the mobile phase consisting of methanol-2% acetic acid in a gradient manner at a flow rate of 1.0 ml .min-1. The detection wavelength was set at 254 nm while the column temperature was at 30 ℃. Results A good linear correlation was observed within the range of 0. 010 92 - 0. 4368 μg for chlorogenic acid （ r = 0. 9999） , 0. 010 20 - 0. 4080 μg for geniposide （ r = 0. 9999） and 0. 042 56 - 1. 7024 μg for baicalin （ r = 0.9999）. The average recovery was 99.97 % with an RSD of 1.57% for chlorogenic acid,99.02% with an RSD of 1.42% for geniposide, and 98.70% with an RSD of 1.52% for baicalin. Conclusion This method is quick, accurate and specific, which can be used for quality evalua- tion of Qingre Jiedu soft capsules.
