摘要
目的:建立一种应用气相色谱-氢火焰离子化检测器定量测定食用油中胆固醇含量的简便方法。方法:样品先经强碱皂化,再用Silyl-991衍生试剂进行衍生净化,然后用正己烷做为提取液,加水除杂,最后利用气相色谱仪的毛细管色谱柱和氢火焰离子化检测器对提取液进行分离和检测。结果:利用该方法,胆固醇与杂质可以得到良好分离.在0.50μg/ml~50.00μg/ml的线性范围内,相关系数R2=0.9998,最低检出量为0.83 ng,当样品添加浓度为0.50 mg/kg~10.00 mg/kg时,方法回收率范围在88.68%~97.40%之间。结论:该方法操作简便,灵敏度高,重现性和选择性好,分离效果良好,最低检出量和回收率均符合微量分析的要求,是检测食用油中胆固醇的快速有效的定性、定量方法。
Objective :To develop a method for cholesterol determination in edible oil by gas chromatography - flame ionization detector( GC -FID). Methods: The cholesterol in edible oil was saponified by alkali solution, and derived from Silyl - 991, then directly extracted by n - hexane and removed impurity by water, finally detected by gas chromatography- flame ionization detector (GC - FID). Results: The detection limit of the method was 0. 83 ng, the recoveries of 88.68 -97.40% were obtained, when dopant concentration was from 0.50 mg/kg to 10.00 mg/kg, RSD was 7.72%. Conclusion : The method for determination of cholesterol in edible oil by GC - FID was high sensitive, easy to operate. It can be used in research of the cholesterol in edible oil.
