1型环氧化酶介导合成的前列环素同步激活小鼠动脉前列环素与血栓烷素受体

摘要 目的:探讨1型环氧化酶(COX-1)是否通过合成前列环素(PGI2)介导小鼠动脉血管舒张以及前列环素(IP)、血栓烷素(TP)受体可能发挥的作用。方法:分离C57BL/6和COX-1基因敲除(COX-1-/-)小鼠肠系膜动脉;张力实验检测COX-1底物花生四烯酸(AA)诱发的血管舒缩反应;高效液相色谱-质谱联用法检测体外肠系膜动脉及血浆中PGI2代谢物6-keto-PGF1α水平。结果:IP和TP受体拮抗剂能分别抑制和增强AA诱导的血管内皮舒张作用。COX-1-/-或抑制剂能消除离体组织6-keto-PGF1α产生,减弱AA诱发的血管舒张或收缩效应。结论:COX-1通过内膜依赖的PGI2合成介导肠系膜动脉的舒张。功能相反的IP和TP受体可同时介导PGI2的血管舒缩反应,IP介导的舒张活性可被TP介导的收缩活性抵消。
