摘要
目的:通过不同时间运动训练对小鼠骨骼肌ERRα表达的研究,探讨耐力训练对骨骼肌内ERRα的累积性应答特征,从而为运动训练对骨骼肌能量代谢信号的调节机制提供理论依据。方法:C57BL/6J野生小鼠,按照运动天数,随机分为7、14、28 d组,每组随机分为2组:安静对照组(C)和耐力训练组(T),另设0 d安静对照组,共7组。Western blot法测定骨骼肌ERRα的总蛋白及核蛋白含量,实时荧光定量PCR测ERRαmRNA含量。结果:1)运动组与安静对照组相比,总蛋白ERRα表达量无显著性差异,而核蛋白ERRα与ERRαmRNA则在14 d与28 d训练组有显著性升高;2)运动组随着训练天数的增加,ERRα的总蛋白与核蛋白表达量呈上升的趋势。结论:1)运动14及28 d可明显提高小鼠骨骼肌核蛋白ERRα以及mRNA的表达;2)随运动时间延长,小鼠骨骼肌ERRα的总蛋白与核蛋白表达累积性增加。
Objective: By the research of effect of training with different time-span on ERRα of mice skeletal muscle,the paper discusses the cumulative response characteristics of endurance training to ERRα in the skeletal muscle so as to provide theoretical basis for the regulation mechanism of sports training on the skeletal muscle energy metabolism.Methods: In this study,C57BL / 6J wide mice were divided into 4 groups randomly based on the different days(0,7,14 and 28),and 7,14 and 28 groups were divided into control and training groups randomly.There were totally seven groups.ERRαmRNA and total,nuclear ERRα protein levels in the skeletal muscle were measured by Real-time PCR and Western blotting respectively.Results: That there were no significant differences in its total protein between training and control group,but ERRα mRNA and the nuclear ERRα protein was much higher than those in the control group during 14 and 28 days.Additionally,the total and nuclear ERRα protein was increased with training days.These findings demonstrate that 14 and 28 days’training can increase ERRα mRNA and the nuclear ERRα protein level,and ERRα protein can be cumulative increased with training days.
