23种中国兜兰属植物亲缘关系的ISSR分析

Analysis of Phylogenetic Relationship of 23 Paphiopedilum Species in China With ISSR Markers

利用ISSR分子标记技术对23种中国兜兰属植物进行种间亲缘关系研究.研究发现:从50条引物中筛选出6条引物进行扩增,共扩增出136条带,其中多态性条带136条,占总条带数的100%,平均每条引物扩增出的DNA条带数为22.7条;根据ISSR扩增结果,采用NTSYS-pc2.10e分析软件进行遗传相似性系数分析,发现23种兜兰的遗传相似性系数在0.145 5～0.780 5之间,平均遗传相似性系数为0.383 9;UPGMA聚类分析发现,在相似性系数为0.292时,23种兜兰被划分为3个类群,即兜兰亚属(Paphiopedilum)、宽瓣亚属(Brachypetalum)和小萼亚属(Parvisepalum),与形态学系统分类结果完全一致.

Inter-simple sequence repeat （ISSR） molecular markers were used to analyze the interspecific phylogenetic relationships of 2a Paphiopedilurn species in China. From 50 primers 6 were screened for amplification, and a total of la6 bands v;,ere generated, all being polymorphic. The average number of DNA fragments produced by each primer was 22.7. Calculated with the software NTSYSpc2.10e, the ge- netic similarity coefficient of the 23 Paphiopedilum species studied ranged from 0. 145 5 to 0. 780 5, avera- ging 0. 383 9. UPGMA cluster analysis divided the 23 Paphiopedilum species into three major groups （Paphiopedilurn, Brachypetalurn and Parvisepalum） at the similarity coefficient of 0. 292, a result corn- pletely consistent with the traditional morphological classification.
