不同鹅种MSTN mRNA的表达特异性及其与体重的相关性分析 被引量:6

摘要 本试验选择生长速度差异较大的太湖鹅和皖西白鹅为试验模型,首次对不同品种鹅胚胎期和生长早期胸肌和腿肌中MSTN mRNA的表达规律及其与体重的相关性进行了比较研究。采用多重荧光竞争性PCR方法研究鹅23、25、27、29胚龄和0、2、4、6、8、10、12周龄胸肌和腿肌中MSTN mRNA的表达规律。研究发现,该基因在23胚龄已有表达。MSTN mRNA在胸肌中两个鹅种的表达趋势一致,在腿肌中两个鹅种的表达趋势不同,但峰值都出现在12周龄。太湖鹅、皖西白鹅体重与胸肌MSTN mRNA的表达呈极显著正相关,提示当促进肌肉生长的基因表达上调时为了避免肌肉细胞的过分肥大MSTN mRNA表达也同时上调。配对T检验发现,两鹅种腿肌中MSTN mRNA的平均表达量均显著高于胸肌,与肌肉增重情况相反,验证了MSTN基因抑制肌肉生长的作用。 The expression of MSTN mRNA in breast and leg muscle was analyzed at the embryonic day(ED) 23,25,27,29 and the age of 0,2,4,6,8,10,12 weeks of Taihu goose and Wanxi goose with Multiplex Competitive Fluorescent-PCR method,and the correlation between expression of MSTN and body weight was also analyzed in this study.The result showed that goose MSTN mRNA has been detected at ED23,and the expression patterns of MSTN mRNA levels in breast muscle of the two goose breeds were relatively consistent during development,but they were different in leg muscle,MSTN mRNA levels in both tissues increased to peak at 12 weeks old.There is a positive correlation between MSTN mRNA levels in breast muscle and body weight of the two breeds.The result indicated that increased up-regulated expression of MSTN maybe inhibit muscle hypertrophy,and muscle regulation factors are in a dynamic balance during muscle development.In contrast to muscle weight gain,paired T-test found that the average expression of leg MSTN mRNA in two geese breeds were significantly higher than the pectoral muscle,which verified that MSTN gene has played a negative regulatory role in muscle growth.These confirmed that MSTN had obvious effect on muscle growth inhibition.
