摘要
目的探讨变应性鼻炎(allergicrhinitis,AR)患者外周血中18T细胞、调节性T细胞(regulatory Tcells,Treg)、白细胞介素(interleukin,IL)17及转化生长因子β1(transforminggrowthfactor β1,TGF—β1)的表达及临床意义。方法2012年3—7月,收集32例AR患者(AR组)和20例健康志愿者(对照组)的外周血,采用流式细胞术检测外周血中18T细胞和Treg细胞比例;酶联免疫吸附试验检测血清中IL-17、TGF-β1的水平,并分析它们之间的相关性。采用SPSS16.0软件进行数据分析。结果叮8T细胞百分比在AR组和对照组分别为(13.30±8.62)%、(5.18±1.86)%,差异有统计学意义(t=5.18,P〈0.01);Treg细胞百分比分别为(1.75±0.56)%、(4.76±1.74)%,差异有统计学意义(t=7.46,P〈0.01)。AR组血清中IL-17的表达水平为(668.55±45.15)pg/ml,高于对照组的(573.53±17.42)pg/ml,差异有统计学意义(t=8.97,P〈0.01);AR组TGF-131的表达水平为(0.34±0.04)pg/m1,低于对照组的(0.49±0.06)0g/ml,差异有统计学意义(t=9.51,P〈0.01)。AR组患者外周血中16T细胞百分比与Treg细胞百分比呈负相关趋势(r=-0.561,P〈0.01),18T细胞百分比与IL-17呈正相关趋势(r=0.469,P〈0.01),18T细胞百分比与TGF—β1呈负相关趋势(r=-0.622,P〈0.01),Treg细胞百分比与TGF—B1呈正相关(r=0.738,P〈0.01),IL-17表达水平与Treg细胞百分比、TGF-β1无相关性(r值分别为-0.111、-0.196,P〉0.05)。结论AR患者外周血中18T细胞和Treg细胞之间存在免疫平衡失调,18T细胞可能参与了IL-17的分泌,这可能在AR的发病和发展过程中发挥重要作用。
Objective To investigate the expression of peripheral blood γδ T cells/CD4+ CD25+ regulatory T cells(Treg) and cytokines interleukin 17-(IL-17) and transforming growth factor β1 (TGF-I31) in patients with allergic rhinitis. Methods From March 2012 to July 2012, 32 patients with allergic rhinitis (AR group) and 20 healthy control subjects (control group) were collected. The expression of peripheral blood γδ T cells/Treg cells were measured by flow cytometry and the levels of IL-17 and TGF-131 were evaluated by ELISA. SPSS 16. 0 software was used to analyze the data. Results The percentages of γδ cells in AR group were (13.30±8. 62)%, which was significantly higher (t = 5.18, P 〈 0. 01 ) than those in control group (5.18± 1.86)%. The percentages of Treg cells in AR group were (1.75 ±0.56)% , which were significantly lower ( t = 7.46, P 〈 0. 01 ) than those in control group (4. 76 ± 1.74 ) %. The IL-17 levels in AR group were (668. 55 ±45. 15) pg/ml, which were also significantly higher (t = 8.97, P 〈0. 01 ) than those in control group (573.53 ±17.42) pg/ml. The TGF-131 levels in AR group were (0. 34 ±0. 04) pg,/ml, which were also significantly lower (t =9. 51, P 〈0. 01 ) than those in control group(0.49 - 0. 06) pg/ml. There was a negative correlation between the percentages of -y8 T cells and Treg cells ( r = - 0. 561, P 〈 0. 01 ). There was a negative correlation between the percentages of γδ T cells and TGF-β1 levels (r = -0. 622, P 〈 0. 01 ). A positive correlation was shown between the percentages of γδT cells and IL-17 levels in AR ( r = 0. 469, P 〈 0. 01 ). A positive correlation was shown between the percentages of Treg cells and TGF-{M levels in AR ( r = 0. 738, P 〈 0. 01 ). There was no correlation between IL-17 levels and the percentages of Treg cells or TGF-β1 levels ( r value was - 0. 111, - 0. 196, all P 〉 0.05 ). Conclusion There are imbalances of γδT and Treg cells in peripheral blood of patients with allergic rhinitis. ~/8 T cells may be the main cell to produce IL-17, which may play an important role in allergic rhinitis.
