咖啡酸对白细胞及血小板减少症小鼠外周血血小板膜糖蛋白表达水平的影响 被引量:3

摘要 目的:观察不同剂量咖啡酸对白细胞及血小板减少症模型小鼠外周血血小板膜糖蛋白CD62p、CD41及CD61表达水平的影响。方法:采用阿糖胞苷制备白细胞及血小板减少症小鼠模型,同时每天一次灌胃给予低(100mg/kg)、中(150mg/kg)、高(200mg/kg)不同剂量咖啡酸,阴性对照组同法给予等量溶媒0.5%羧甲基纤维素钠,阳性对照组给予重组白细胞介素-11。结果:给药第10天,咖啡酸中剂量及高剂量组血小板膜糖蛋白CD62p的表达水平明显高于正常及模型阴性对照组;各模型组血小板膜糖蛋白CD41及CD61表达水平较正常明显降低;与阴性对照组比较,阳性对照组及咖啡酸各剂量组对CD41及CD61表达无明显影响。结论:咖啡酸中剂量及高剂组能明显升高模型小鼠血小板膜表面的CD62p表达水平,活化血小板,对于启动和扩大血栓形成,从而增强凝血功能具有重要意义。 To explore the effects of different doses of caffeic acid (CA) on expression of glucoproteins on the platelet membrane in leu- copenia and thrombocytopenia mice. Methods: Myelosuppressed model mice were prepared by chemotherapy drug cytosine arabinoside ( Ara- C), and treated with low-dose CA (100 mg/kg), intermediate-dose CA (150 mg/kg), and high-dose CA (200 mg/kg) by garage mean- while, 0.5% earboxymethylceilulose sodium (vehicle) was used as negative controls, once daily by gavage, and interleukin-11 was used as positive control once daily by subcutaneous injection. Results: The expressions of CD62p on platelet membrane in model mice treated with intermediate-dose and high-dose CA were significantly higher than that in normal group and negative control group on day 10. The expressions of CIM41/CD61 in all of the Ara-C-treated groups were significantly lower than that in normal group, and there were no significant differences in positive control group and groups treated with different dose of CA, compared with negative control group. Conclusion: Treatment with in- termediate-dose and high-dose of CA could increase significantly expression of CD62p on platelet membrane in model mice to activate plate- lets, and play an important role in initiating and enlarging thrombus to strengthen coagulative function.
