莲心碱对大鼠局灶性脑缺血/再灌注损伤的抗氧化作用 被引量:5

Study on antioxidation of liensinine on focal cerebral ischemia-reperfusion injury in rats

摘要 目的:探讨莲心碱对大鼠局灶性脑缺血/再灌注损伤的抗氧化作用。方法:40只大鼠随机分为5组,对照组、模型组组、莲心碱低、中、高剂量(舌静脉注射莲心碱1.5mg/kg、3mg/kg、6mg/kg),每组8只。采用线栓法阻塞大鼠大脑中动脉2h/再灌注24h制备脑缺血再灌注模型,观察莲心碱对大鼠局灶性脑缺血2 h再灌注24 h后神经功能障碍,脑水肿及脑梗塞范围以及脑组织能量物质ATP、葡萄糖无氧酵解产物以及体内自由基清除剂SOD、GSH和脂质过氧化产物MDA等生化物质的影响。结果:与对照组神经功能评分(0.00±0.00)比较,模型组神经功能评分(3.69±0.85)显著增高;与模型组比较,莲心碱中剂量组神经功能评分为(2.05±1.23)显著降低,莲心碱高剂量组神经功能评分(1.96±1.21)显著降低(P<0.01)。与对照组脑缺血体积比较,模型组脑缺血体积(28.91±3.71)%显著增加;而莲心碱低剂量组、中剂量组、高剂量组脑缺血体积分别为(25.36±3.52)%、(10.33±2.87)%、(9.91±2.61)%。与模型组比较,莲心碱低剂量组脑缺血体积无明显缩小;莲心碱中、高剂量组脑缺血体积明显缩小。与对照组脑组织含水量(77.33±4.34)比较,模型组脑组织含水量为(86.55±5.45)%明显高于假手术组(P<0.01),而莲心碱低剂量组、中剂量组、高剂量组脑组织含水量分别为(83.17±4.68)%、(80.03±4.59)%、(79.12±4.45)%;与模型组相比,莲心碱中剂量组、高剂量组脑组织含水量明显减少。与对照组大鼠血清SOD活性(61.44±15.23)、GSH-Px活性(225.91±8.81)、MDA含量(5.3±0.80)比较,模型组大鼠血清SOD活性(28.38±14.98)、GSH-Px活性(176.87±8.12)均显著降低(P<0.01),MDA含量(8.2±0.90)显著升高(P<0.01);与模型组比较,莲心碱中剂量组大鼠血清中SOD活性(54.23±18.32)显著升高,莲心碱高剂量组大鼠血清中SOD活性(57.12±17.51)显著升高(P<0.01);莲心碱中、高剂量组大鼠血清中GSH-Px活性为(199.61±12.67)、(206.13±14.16)均显著升高(P<0.01);莲心碱中、高剂量组大鼠血清中MDA含量为(6.7±0.80)、(6.1±0.81)显著降低(P<0.01)。与对照组比较,模型组脑组织内ATP含量(1.45±0.16)明显降低,脑组织内LA含量(8.84±0.45)明显升高。与模型组比较,莲心碱中剂量组脑组织内ATP含量为(1.76±0.22)显著升高,莲心碱高剂量组脑组织内ATP含量为(1.89±0.23)显著升高;莲心碱中、高剂量组脑组织内LA含量为(6.14±0.34)、(5.89±0.31)明显降低。表明莲心碱能显著改善大鼠的神经系统损伤症状,缩小脑缺血体积,增强血清SOD和GSH-Px活性、减少MDA含量,大鼠脑组织内ATP含量显著升高,而LA含量明显降低。结论:莲心碱能显著提高局灶性脑缺血/再灌注大鼠的抗氧化能力。 To study the antioxidative effect of lieusinine on focal cerebral ischemia-reperfusion injury in rats. Methods: 40 SD rats were divided into 5 groups : The sham group, the model group, the high dose of liensinine ( 6mg/kg), the moderate dose of liensinine ( 3mg/ kg) ,the low dose of liensinine(1.5mg/kg). The right middle cerebral artery of the rat was occluded by inserting a thread through internal ca- rotid artery for 2h, and then reperfused for 24h. Liensinine was injected after ischemia. After 24h, Use the Zea Longa grade point standard to valuate the hurt degree of the nervous system, the TYC dies calculate volume ratio of the cerebral ischemia. The activities of super-oxide dismutase (SOD), glutathione peroxidase (GSH-Px) and the contents of malonic dialdehyde (MDA) in serum were assayed respectively. The levels of ATP and LA in brain were tested. Results: Compared with the sham group, the nervous system hurt symptom (3.69 ± 0. 85 ) was significant increased in the ischemia group. Compared with the isehemia group, the nervous system hurt symptom in the low dose of liensi- nine was (2.83 ± 1.31 ), had no marked effect. The nervous system hurt symptom in the moderate dose of liensinine was (2.05 ± 1.23 ), the nervous system hurt symptom in the high dose of liensinine was ( 1.96 ± 1.21 ), was significant decreased. Compared with the sham group, the cerebral ischemia volume ( 28.91 ±3.71 )% was significant increased in the ischemia group. Compared with the ischemia group, The cerebral ischemia volume in the low dose of lieusinine was ( 25.36 ±3.52) %, had no marked effect. The cerebral iachemia volume in the moderate dose of liensinine was( 10.33 ± 2.87 )%, the cerebral ischemia volume in the high dose of liensinine was( 9.91 ± 2.61 )%, had significant decreased. Compared with the sham group,the cerebral water content was ( 86.55 ± 5.45) % ,was significant increased in the ischemia group. Compared with the ischemia group, the cerebral water content in the low dose of liensinine was (83.17 ± 4.68) %, had no marked effect. The cerebral water content in the moderate dose of liensinine was ( 80.03 ± 4.59) % ,the cerebral water content in the high dose of liensinine was (79.12 ± 4.45 ) %, was significant decreased. Compared with the sham group, the activities of SOD was (28.38 ± 14. 98 ), and GSH-Px was (176.87 ± 8.12) ,was significant decreased in the ischemia group. And MDA was (8.2 ±0.90), was significant in- creased in the ischemia group. Compared with the iachemia group, The activities of SOD in the moderate dose and the high dose of liensinine was(54.23 ± 18.32), (57.12 ± 17.51 ), was significant increased. The activities of GSH-Px in the moderate dose and the high dose of liensinine was( 199.61 ± 12.67), (206.13 ± 14.16) ,was significant increased. The content of MDA in the moderate dose and the high dose of lieusinine was (6.7 ± 0.80), (6.1 ± 0.81 ), was significant decreased. Compared with the sham group, the content of ATP (1.45 ± 0.16) was significant decreased and the content of LA( 8.84 ± 0.45 ) was significant increased in the isehemia group. Compared with the ischemia group, the content of ATP in the moderate dose and the high dose of liensinine was ( 1.76 ± 0.22), ( 1.89 ± 0.23 ), was significant increased. The content of LA in the moderate dose and the high dose of liensinine was(6.14 ±0.34), (5.89 ±0.31 ) ,was significant decreased. These results showed lieusinine could improved the nervous system hurt symptom, reduced the cerebral ischemia volume smaller in the lieusinin group. The activities of SOD and GSH-Px in surum were all increased and the contents of MDA got high reduction. The levels of ATP were increased and LA were decreased. Conclusion: The liensinine can significantly increase the antioxidative effect on focal cerebral ischemia- reperfusion injury in rats.
