附子与半夏预处理对大鼠心肌缺血再灌注损伤细胞凋亡及SOD、MDA的影响 被引量:21

Effects of Monkshood and Pinellia preconditioning on SOD,MDA and apoptosis in rats with myocardial ischemia-reperfusion injury

摘要 目的:观察含附子、半夏单用及配伍应用对大鼠心肌缺血再灌注损伤的保护作用,探讨附子与半夏配伍用于特定病理状态的可行性。方法:大鼠随机分为5组:假手术组、模型对照组、附子组、半夏组、附子+半夏组。每日以灌胃法给予假手术组、模型对照组为等容量生理盐水,附子组为白附片单煎剂,半夏组为姜半夏单煎剂,附子+半夏组为白附片+姜半夏混合煎剂,每日给药剂量为2.0(生药)g/kg体质量。除假手术组外,其他各组结扎冠状动脉左前降支,使心肌缺血30min,再灌注120min,检测血清超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量;TUNEL法检测各组心肌细胞凋亡指数;RT-PCR法测bcl-2、bax的mRNA表达。结果:按2.0(生药)g/kg体质量给药时,附子+半夏组的MDA释放量比模型对照组、附子组、半夏组均有所减少,SOD含量明显增加;附子+半夏组较模型对照组、附子组、半夏组凋亡指数均降低;附子组、半夏组与模型对照组相比,抑凋亡基因bcl-2 mRNA、促凋亡基因bax mRNA表达水平均无明显变化,附子+半夏组与模型对照组相比,bcl-2 mRNA表达水平上升,bax mRNA表达水平下降,bcl-2/bax比值升高。结论:附子与半夏配伍可能通过抗自由基和减少细胞凋亡,对心肌缺血再灌注损伤产生保护作用。 To investigate the effects of Monkshood and Pinellia on myocardial ischemia-reperfusion injury in rats, and to explore the feasibility of Monkshood cooperating with Pinellia for the specific pathophysiological conditions. Methods: 50 healthy male Wistar rats were randomly divided into five groups ( 10 for each group). Monkshood with myocardial ischemia-reperfusion injury (IR) group, Pinellia with IRgroup and Monkshood & Pinellia with IR group was intragastrically administrated respectly with Monkshood, Pinellia, Monkshood & Pinellia with 0.25 g/100 g body weight concentration for consecutive 14 days. Sham-operation group and I/R group were received normal sodium. Except sham-operation group, other four groups were given the operation of the left anterior descending branch of coronary arteray ligation, myocardium were ischemia for 30 minutes, reperfusion for 120 minutes. All rats were sacrificed after reperfusion, blood serum and hearts were collected for further analysis. Malondialdehyde (MDA) and Superoxide dismutase (SOD) in blood serum was analyzed, Apoptotic in- dex, apeptotic genes expression in myocardium were investigated. Results: Comparing with I/R group, Monkshood & Pinellia group signifi- cantly suppressed MDA release and myocardial apeptosis, SOD of Monkshood & Pinellia group stepped up. Monkshood cooperating with Pinelliag pretreatment also modulated bcl-2/bax ratio by up-regulating bcl-2 while decreasing bax expression. Conclusion: Monkshood coop- erating with PineUia may protect myocardium from I/R injury via anti-free radical and decreasing myocardial cell apoptosis.
