摘要
目的:研究脑舒胶囊对VD模型大鼠的中枢免疫炎症反应的抑制作用。方法:反复夹闭双侧颈总动脉加硝普钠降压复制VD大鼠模型;Morris水迷宫检测大鼠学习记忆力;放射性免疫法测定血清IL-6含量;ELISA法测定血清sIL-2R含量;免疫组织化学检测TNF-α阳性表达。结果:与假手术组比较,模型组大鼠血清IL-6及sIL-2R含量明显增加,与模型组相比,0.36g/kg、0.72g/kg、1.44g/kg脑舒胶囊均可降低大鼠血清IL-6及sIL-2R含量,具有一定剂量依赖性。光镜下(×400)下,模型组大鼠海马CA1区胞浆及胞膜棕褐色颗粒明显增加,各剂量脑舒胶囊治疗组大鼠海马CA1区胞浆及胞膜棕褐色颗粒明显减少;与假手术组比较,模型组TNF-α阳性反应的平均光密度(ID)均明显降低,各治疗组TNF-α阳性反应的ID均下降,且有一定剂量依赖性。结论:脑舒胶囊对VD大鼠中枢免疫炎症反应有较好抑制作用。
To research the inhibition of Naoshu Capsule on central immune inflamation in rats with VD. Method: VD rat model was in- duced by closure both common carotid arterys and reperfusion several times, acuity of learning and memory of rats was measured with Morris water maz; serumal IL-6 was detected by Radio Immunoassay and serumal sIL-2R were detected by ELISA, expression of TNF-α was ob- served by immunohistoehemistry. Results: Compared with sham operation group, serumal IL-6 and sIL-2R increased in model group( P 〈0. 01 ). Compared with model group, Naoshu Capsule (0.36, 0.72, 1.44g/kg) decreased serumal IL-6 and sIL-2R( P 〈 0.05, P 〈 0.01 ). In light microscope ( × 400), in model group, brown granules in endochylema and membrane of VD rats' hippocampus CA1 domain increased, and that decreased obviously in Naoshu Capsule groups. Compared with sham operation group, the ID of TNF-α of model group decreased ob- viously( P 〈0.01 ), and the ID of TNF-αof Naoshu Capsule groups decreased. Dosage dependence was observed in Naoshu Capsule group. Conclusion: Naoshu Capsule can inhibite central immune inflamation of VD rats.
