类叶升麻苷抗H2O2诱导的PC12细胞氧化损伤的保护作用 被引量:13

摘要 目的:类叶升麻苷对H2O2诱导的PC12细胞损伤的影响。方法:以300μmol/L的H2O2处理PC12细胞,建立氧化损伤模型,并以5、10、20、40、60、80、100μg/ml的类叶升麻苷拮抗其作用。通过倒置显微镜观察细胞形态,采用MTT比色法检测细胞存活率,酶标法试剂盒测定LDH渗漏率,TBA法试剂盒测定胞内胞外MDA含量,WST法试剂盒测定胞内胞外SOD活性。结果:类叶升麻苷浓度从10μg/ml起,即可显著改善PC12细胞形态,提高细胞的存活率,减少细胞LDH的渗漏,抑制胞内和胞外MDA的生成,提高胞内和胞外SOD酶活性,且随着给药剂量的增加,其对损伤细胞的改善作用也愈强。结论:类叶升麻苷对H2O2诱导的PC12细胞损伤具有显著保护作用,其机制可能与抗氧化作用有关。 Objective:To investigate the effect of acteoside on Oxidative damage of PC12 induced by H2O2.Methods: Establishment of the PC12 oxidative damage cellular model through 300μmol/L H2O2 treatment.And then antagonize the effect of H2O2 by different concentration(5,10,20,40,60,80,100μg/ml) acteoside.Observed the cell morphology by inverted microscope,determinated the cell survival with MTT,measured the LDH release by enzyme label kit,analysis the MDA contents and SOD activity by TBA kit and WST kit.Results: The concentration of acteoside starting from 10μg/ml could significantly improve the cell morphology and the cell survival,decreased the cell LDH release and it is not only could inhibit the production of intracellular and extracellular MDA,but also could increase the activety of intracellular and extracellular SOD.Moreover,the improving effect show stronger with the drug concentration increased.Conclusion: The acteoside has significant protective effect on the PC12 oxidative damage cellular model induced by H2O2,and it's protective mechanism probably relate to the antioxidant.
