摘要
目的:研究大黄苷元对大鼠肝微粒体CYP450酶活性的影响。方法:将大鼠随机分组,对照组给予0.5%羧甲基纤维素钠(CMC-Na),诱导剂组苯巴比妥钠75.0mg/kg、地塞米松80.0mg/kg、β-萘黄酮75.0mg/kg,抑制剂组酮康唑50.0mg/kg,大黄苷元高、中、低剂量组分别为57.6mg/kg、28.8 mg/kg、14.4 mg/kg,计算各组肝微粒体肝脏指数,测定各组肝微粒体蛋白含量,用CO还原差示光谱法(Omura法)测定CYP450酶的含量,评价酶活性。结果:与CMC组及酮康唑组比较,大黄苷元CYP450酶活性升高;与苯巴比妥钠组、地塞米松组及β-萘黄酮组比较,CYP450酶活性低。结论:大黄苷元对大鼠肝微粒体CYP450酶的活性有诱导作用,且有随给药剂量的增加而增强的趋势。
Objective:Study the anthraquinones from Radix et Rhizoma Rhei on activity of liver microsomal CYP450 in rats by enzyme assays.Methods: SD rats are randomly grouped,control group was given 0.5% sodium carboxymethyl cellulose(CMC-Na),induction agent group phenobarbital sodium(PB) 75.0mg/kg,dexamethasone(DEX) 80.0mg/kg,beta-naphthoflavone(β-NF) 75.0mg/kg,inhibitor group ketoconazole 50.0mg/kg,Anthraquinones high,medium and low-dose group were 57.6mg/kg,28.8mg/kg,14.4mg/kg,calculate the index of the liver microsome,determination of liver microsomal protein content,reduction by CO difference spectra France(Omura France) determination of the content of the liver microsomal CYP450 enzyme content,and evaluation of enzyme activity.Results: Compared with CMC group and the ketoconazole group,anthraquinones CYP450 enzyme activity increased(P 0.01);compared with the PB group,DEX group and β-NF group,the CYP450 activity decreased(P 0.05).Conclusion: The anthraquinones from Radix et Rhizoma Rhei could induce rat liver microsomal cytochrome P450 activity and enhanced with the increase of the dose trend.
