紫草对重症急性胰腺炎肾组织的影响 被引量:4

Study on the impact of Arnebia euchroma on renal tissue protection in rat severe acute pancreatitis

摘要 目的:观察紫草对大鼠重症急性胰腺炎(SAP)肾组织热休克蛋白70(HSP70)表达及血清SOD、MDA、NO、肌酐和尿素氮的影响,探讨其作用机制。方法:大鼠随机分为假手术组、模型组、紫草3g/kg和紫草1g/kg组,检测术后24h和72h肾HSP70的表达及血清SOD、MDA、NO和肌酐和尿素氮变化。结果:紫草的起始剂量为1g/kg,其剂量范围在1~3g/kg,紫草3g/kg组和紫草1g/kg组死亡率明显低于模型组;紫草3g/kg组和紫草1g/kg组SOD和NO活性较高而MDA、肌酐和尿素氮水平较低,与模型组相比P<0.01;术后24h肾组织HSP 70表达比72h多(P<0.05),假手术组表达较少,模型组表达明显增多,紫草3g/kg组和紫草1g/kg组表达明显下调,与模型组比较(P<0.01),相关分析发现,肾组织HSP 70表达与血清MDA、肌酐和尿素氮呈正相关,与SOD和NO呈负相关。结论:紫草能降低SAP大鼠死亡率,提高血清SOD和NO活性、抑制MDA、肌酐和尿素氮含量、下调肾组织HSP70表达,减轻肾损伤,保护肾功能。 Objective:To observe the effect of Arnebia euchroma on the SOD(superoxide dismutase) and MDA(malondialdehyde) and NO(Nitric oxide) and creatinine(Cr) and urea nitrogen(BUN) in blood serum of rat severe acute pancreatitis(SAP),kidney tissue optical microscope the morphology of the change and on renal injury heat shock protein 70(HSP70) expression,then investigate its role of on the intervened action and mechanism involved.Methods: Wistar rats were randomized into four groups,the sham operated control group(group A only laparotomy) 、SAP group(group B model group) and Arnebia euchroma erythrorhizon treatment group(group C and D group),Postoperative 24h and 72h after detected serum SOD、MDA and NO and Cr and BUN detected,taken kidney tissues immunohistochemical method in expression of renal tissue's HSP70 after operation.Results: Death rate of C and D group was lower than B group(P 0.01);the activity of SOD and NO was higher and MDA and Cr and BUN expression was lower in the group of Arnebia euchroma treatment than control(P 0.01) Group A HSP70 expression in the kidney tissues less,group B in the kidney tissues HSP70 express significantly increased,group C and D group HSP70 express clear cut,and the comparison group B,significant difference(P 0.01).Correlation analysis found that renal tissue HSP 70 expression and serum MDA,creatinine and urea nitrogen are positively related,and SOD and NO negatively related.By light microscopy visible: group A rat bilateral kidneys no significant pathological change;Group B renal tubular epithelial cell necrosis fall off,renal tubular edema;Group C and D group of renal tubular edema degree is group B significantly reduce,did not see the renal tubular epithelial cell necrosis.Conclusions: Arnebia euchroma could significantly SAP reduce mortality rate,increased the activity of SOD and increase NO level,decreased the generation of MDA and Cr and BUN,cut in the kidney tissues HSP70 expression,reduce the SAP of the kidney damage in rats.
