摘要
研究了经 TAA慢性处理而诱发的肝纤维化期间枯否细胞中凋亡基因和细胞周期基因的表达。按照肝脏损伤的程度把实验动物 Wistar大白鼠分为 2组 ( TAA-1和 TAA-2 )。经核 Run-off转录分析表明 ,在2个试验组的枯否细胞中 ,总的 RNA合成率下降约 2 0 %。在 TAA-1的枯否细胞中 ,bcl-2和 bcl-x的 m RNA合成增加了 4倍 ,而 cyclin E在肝损伤的前期有所降低。在 TAA-2中 ,bad、cyclin B和 cyclin D1的 m RNA合成稍微降低 ,而 bax、bcl-2和 cyclin E的 m RNA合成分别明显升高 ,bcl-x和 cyclin A的 m RNA合成分别增加了 2 .5倍和 4 .5倍。这些表明了在
In this paper, the expression of apoptosis and cyclin genes was studied in isolated Kupffer cells (KC) of chronically injured livers of Wistar rats. Experimental animals by oral treatment of TAA for 3~6 months were divided into two groups (TAA 1 and TAA 2) according to the extent of liver injury. The analysis of nuclear run off transcription assays showed that the rate of incorporation of 32 P UTP into total RNA by isolated nuclei of KC decreased significantly by about 20% in both experimental groups. The synthesis of mRNA of bcl 2 and bcl x increased by 4 fold in SEC at the early stage of liver injury, of the mRNA of cyclin E was moderately decreased . At the late stage of TAA induced fibrogenesis (TAA 2), the synthesis of mRNA of bad, cyclin B and cyclin D1 in KC slightly decreased while the bax,bci 2 and mRNA of the cycllin E increased remarkably. The synthesis in mRNA of bcl x and cyclin A increased by 2.5 fold and 4.4 fold, respectively. The findings clearly indicated the regulation of the proliferative activity and apoptotic processes in KC induced by TAA during TAA hepatic fibrogenesis had changed.
