猪PBMC产生IL-2最适条件的探讨

Appropriate Condition for Pig PBMC to Producing IL 2

应用 MTT法 ,通过正交试验对影响体外培养的猪 PBMC分泌 IL-2的 4个因素进行了综合分析 ,证明培养时间、PHA-P和细胞密度对 IL-2分泌有极显著或显著影响 ( P<0 .0 1或 P<0 .0 5) ,NCS在 2 .5%～2 0 %范围内 ,影响不显著 ( P>0 .0 5)。体外诱导猪 PBMC分泌 IL-2的最适条件为细胞密度 5× 1 0 6个 /ml,PHA-P终浓度为 5mg/L,NCS浓度为 2 .5%～ 2 0 %,于 3 7℃、5%CO2 、饱和湿度条件下培养 2 4

By MTT colorimetric assay, and based on orthoganal experimental design, 4 factors which affect IL 2 of pig PBMC in vitro were analyzed. The results showed that the culture time, concentration of PHA P and density of cells had significant effect on IL 2 ( P <0.01 or P <0.05), within the scope of 2.5% ～20%, it had slight effect . The appropriate condition for inducing IL 2 of the pig PBMC was: cells density was 5×10 6/ml, the concentation of PHA P 5 mg/L,the concentration of NCS 2.5%～20% at 37℃, 5% CO 2 for 24 h.
