摘要
[目的]观察丹酚酸对房颤大鼠外周血基质金属蛋白酶,组织型基质金属蛋白酶抑制因子(MMP-2/TIMP-2)、肿瘤坏死因子α(TNF-α)表达影响。[方法]使用随机平行对照方法,将40只清洁级SD大鼠随机取出10只作为空白组。另外30只采用乙酞胆碱-氯化钙(Ach-Cacl2)混合液尾静脉注射法复制房颤动物模型,连续4周,心电图出现典型房颤为造模成功。由于大鼠部分死亡,将24只造模成功的实验大鼠按区组随机方法随机模型组、维拉帕米组、丹酚酸组,8只/组,空白对照组也随机减去2只。造模成功2d后,各组灌胃干预,维拉帕米20mg/kg.d,注射用丹参多酚酸4mg/kg.d,模型组、对照组用等量蒸溜水;1次/d,连续27d,自由饮水,固定饲料。用ELISA法检测血清MMP-2/TIMP-2、TNF-α。[结果]与空白组比较,模型组MMP-2升高(P>0.01),TIMP-2降低(P<0.01);维拉帕米组、丹酚酸组MMP-2均升高(P<0.01,P<0.05),维拉帕米组、丹酚酸组TIMP-2均降低不明显(P>0.05);模型组,丹酚酸组、维拉帕米组TNF-α均增高(P<0.01,P<0.05)。[结论]丹酚酸可改善房颤大鼠心房肌细胞重构过程的损伤程度,从而治疗房颤。
[Objective]To observe Dan phenolic acids on atrial fibrillation rats peripheral blood matrix metalloproteinases,tissue matrix metalloproteinases inhibitory factor(MMP-2 / TIMP-2),tumor necrosis factor alpha(TNF alpha) expression.[Methods]The calcium chloride-acetylcholine mixture made mold,40 SD rats were randomly remove 10 as blank group.30 acetylcholine-calcium chloride(Ach-CaCl2) copy mixture tail vein injection animal models of atrial fibrillation for 4 weeks,typical atrial fibrillation in ECG modeling success.As part of death in rats,the 24 successful model in rats model group by block randomisation method random,verapamil group,Salvianolic acid group,8 / group,blank group randomized minus 2.Modeling 2d,verapamil group were given verapamil 20mg / kg d orally;the salvianolic acid treatment group was given Danshen injection of acid 4mg / kg d orally;model group and the control group with an equal amount of distilled water orally.1 times / d,for 4 weeks,free drinking water,given a fixed feed.ELISA assay of serum MMP-2 / TIMP-2,TNF-alpha changes.[Results]The model group TNF alpha,MMP-2 level higher,lower TIMP 2;Dan phenolic acid treatment group,verapamil intervention group were decreased peripheral blood TNF alpha,MMP-2 level,elevated TIMP-2 level,and Dan phenolic acid group effect of verapamil group.[Conclusion]Dan phenolic acids af can be improved through the rat heart muscle cells in the process of reconstruction of damage degree,and treatment of atrial fibrillation.
