The study of human peripheral blood lymphocyte chromosome slides-making techniques

摘要 目的改进人外周血淋巴细胞染色体制片技术,探寻一种更简单方便的滴片方法,提高染色体分析效率。方法抽取正常人外周血,按照本实验室经典方案制备淋巴细胞悬液,采用4种不同滴片方法进行滴片,观察并分析不同滴片方法制备的染色体标本效果(重复性及稳定性)。结果采用干冰片过火法获得的可分析染色体分裂相数目丰富,形态结构好,染色体分散面积大,差异小,一个分裂相交叉重叠的染色体少,染色体分裂相破裂的频率低。结论通过本实验研究发现了一种简单可行的有助于染色体最佳扩散的制片技术,大大提高了染色体分析效率。 Objective: Improving human peripheral blood lymphocyte chromosome slides-making techniques,exploring a simpler and convenient drop slide method,and increasing the chromosome analysis efficiency.Methods: To draw normal peripheral blood,harvest lymph cell suspension according to our laboratory classic protocol,and drop slides using four different methods.To observe and analyze the chromosome slides by different making methods(repeatability and stability).Results: We may get more metaphase chromosomes,better morphosis,larger metaphase area,less chromosome overlaps,or lower frequencies of broken metaphases using dried and iced slides over fire methods..Conclusion: A simple and feasible method for improving chromosome spreading was developed based on our study.
