摘要 目的研究麻风患者皮损中Foxp3+的表达,阐明Foxp3+T细胞与麻风发病的关系。方法麻风患者皮损30例,正常对照组20例,用免疫组化研究麻风患者皮损内Foxp3+细胞分布情况。结果①Foxp3+细胞主要位于麻风皮损真皮层,正常对照皮肤中Foxp3+细胞数量较少或无;②TT和BT型,Foxp3+细胞分布在肉芽肿及其周围。BL型和LL型,Foxp3+细胞散在分布于浸润的巨噬细胞间;③Foxp3+在TT,BT,BL和LL组表达均强于正常组(P<0.001)。BL,LL之间,Foxp3+蛋白表达无统计学差异(P>0.05)。BT组,TT组,Foxp3+蛋白表达均高于LL组,差异无统计学意义(P>0.05);TT组Foxp3+蛋白表达高于BT组,但差异无统计学意义(P>0.05)。结论①Foxp3+Treg细胞是麻风病发病的重要参与者;②提示调节性T细胞和组织细胞之间可能存在相互作用;③实验组Foxp3+Treg淋巴细胞表达高于健康对照组,提示麻风患者细胞免疫功能低下可能与Foxp3+Treg淋巴细胞有关。 Objective To investigate the expression of Foxp3 + Tregs cells in lesions of leprosy, in order to if luminate discuss the role of CD4 + CD25 +Tregs cells in leprosy. Methods The distribution of Foxp3 + cells in 30 patients with leprosy and 20 normal controls were de- tected by immunohistochemistry staining method. Results Foxp3 + cells in leprosy predominantly located in dermis, and it was absent or scant in normal skin. In TF and BT, Foxp3- cells were observed both inside and around the granulomas; In BL and LL, Foxp3-cells were scattered in the infiltrated macrophages. Compared with control, the expression of Foxp3 + significantly increased in TT, BT, BL and LL (P〈0. 001 ). The expression of Foxp3 was not significant difference between BL and LL. The expression of Foxp3- in BT and TT was higher than LL, but there was no significant difference( P〉0. 05 ). The expression of Foxp3 was not significant difference between BL and LL, although Foxp3 expression were increased in TT compared to BT (P〉0. 05 ). Conclusion The Foxp3-treqs cells play an important role in the pathogenesis of leprosy. There would be interaction between CD4+CD25+Foxp3 Tregs and histiocytes. The expression of Foxp3 significantly increased in patients with leprosy compared with control, which indicated that there would be immunosuppressive in leprosy.
