板栗总苞多酚纯化工艺优化及其对细胞活力的影响 被引量:3

Purification Technology of Polyphenols from Castanea Mollissima Blume with Macroporous Absorbent Resin

摘要 比较10种大孔吸附树脂对板栗总苞中多酚的吸附与解吸性能。在静态吸附研究基础上,筛选较好的树脂做动态试验,并比较所得多酚与纯化前多酚对IEC-6细胞在不同条件下增殖能力的影响。结果表明:AB-8大孔吸附树脂纯化板栗总苞多酚的效果最好。上柱液质量浓度1 mg/mL,流速2 mL/min,上柱液体积400 mL,洗脱液乙醇质量分数70%,洗脱流速1 mL/min,洗脱液体积140 mL。在此条件下,多酚的吸附率和解吸率分别为87.2%和97.7%,板栗总苞多酚纯度由47.86%提高到81.67%。板栗总苞多酚对IEC-6细胞增殖有促进作用,且多酚纯化后比纯化前效果明显。 Objective:With the aim to purify polyphenols from Castanea mollissima Blume by macroporous resin,eleven macroporous resins were tested for their static adsorption/desorption performance towards polyphenols.Based on the static adsorption experiments,the suitable macroporous resin was screened to carry out dynamic adsorption experiment,and it was compare that effect of polyphenols impurified/purified on cell proliferation of IEC-6.The results showed that AB-8 was the optimal macroporous resin for purifying polyphenols,the optimum purifying condition was as follows:the mass concentration and the sampling rate were 1 mg/mL and 2 mL/min,respectively.Polyphenol sample volume was 400 mL.Whereas the optimal desorption conditions are found to be a flow rate of 1mL/min using 140 mL of 70% ethanol for elution.The final adsorption rate and desorption rate are 87.2% and 97.7%,respectively.The purity of polyphenols from Castanea mollissima Blume increases from 47.86% to 81.67%.Polyphenols from Castanea mollissima Blume can promote cell proliferation of IEC-6,and polyphenols purified was more obvious effect and impurified.
