摘要
目的观察补气化痰方对心肌梗死大鼠心功能及VEGF表达的影响。方法将72只雄性SD大鼠随机分为治疗组(补气化痰方)、中药对照1组(黄芪)、中药对照2组(瓜蒌薤白半夏汤)、西药对照组(氯沙坦钾)、模型组(仅进行心梗造模)、正常组(假手术组),每组12只。除正常组外,其余各组结扎前降支建立心肌梗死模型,并予相应的措施干预28天。观察比较各组的大鼠生存率、心脏超声射血分数(EF),心肌VEGF mRNA和蛋白表达情况。结果①各组大鼠生存率差异无统计学意义(P>0.05)。②除正常组外,各组心脏EF下降(P<0.05)。复查心脏超声,与模型组比较,治疗组EF差异有统计学意义(P<0.05);术后28 d与术后48 h EF的差值比较,治疗组较模型组、中药对照1组、中药对照2组、西药对照组EF下降缓慢,差异有统计学意义(P<0.05)。③治疗组较模型组、中药对照1组、中药对照2组、西药对照组VEGF mRNA和蛋白的表达增高,差异有统计学意义(P<0.05)。结论补气化痰方能有效提高心肌梗死大鼠心脏EF值,其可能的机制之一是调控缺血心肌VEGF表达,从而改善心脏功能。
Objective To observe the effect of "Buqi Huatan Decoction" on cardiac function and VEGF expression in rats with myocardial infarction.Methods Seventy-two male SD rats were randomly divided into treatment group(Buqi Huatan Decoction),Chinese medicine control group 1(Radix Astragali seu Hedysari),Chinese medicine control group 2(Gualou Xiebai Banxia Decoction),western medicine control group(losartan potassium),model group and normal group,12 rats in each group.The myocardial infarction models were established by ligation the anterior descending branch and intervened for 28 days.The survival rate,48 h,28 d cardiac ultrasound EF and 28d VEGFmRNA expression were observed.Results ① No significant difference was found in survival rate in each group within 28 days(P0.05).② After 48 hours,cardiac ultrasound was detected.Except for normal group,the EF and activity of cardiac anterior wall were decreased.Compared with the normal group,there was a significant difference(P0.05).After 28 days,there was significant difference in EF between the model group and treatment group(P0.05).The EF of the 28 days and 48 hours after operation in treatment group was decreased and significantly different from the model group,Chinese medicine group1,Chinese medicine group2,western medicine group(P0.05).③ The expression of VEGF mRNA at 28 days after operation showed that: the expression of VEGF mRNA in treatment group was increased markedly and different from the model group,Chinese medicine group1,Chinese medicine group2,western medicine control group(P0.05).Conclusion "Buqi Huatan Decoction" can effectively improve the EF value in myocardial infarction rats,and its mechanism may be association with regulation of VEGF expression.
