摘要
文章比较了三种血浆中免疫球蛋白IgG含量及活性的检测方法,分别采用单向免疫扩散法、聚丙烯酰胺凝胶电泳法及高效液相色谱法(即HPLC法)对猪血浆中IgG活性及含量进行检测,同时比较三种方法的可行性与准确性。结果表明,免疫扩散检测结果偏差大,电泳法只能初步对比出IgG含量,故两者均不适合用于猪血浆中IgG含量定量检测;HPLC法操作简便,可对样品中IgG进行定性定量检测,检测结果准确度高,重复性好。
This paper compares three different methods of detecting immunoglobulin G (IgG) from swine plasma. Single immunodiffusion method, polyacrylamide gel electrophoresis method and High-performance liquid chromatography (HPLC) method are used to detect IgG from swine plasma respectively. Simultaneously, feasibility and accuracy are also assessed. The results show that large deviation is always found in single immunodiffusion method. Electrophoresis method can only compare with the level of IgG preliminary. Single immunodiffusion method and polyacrylamide gel electrophoresis methods are not suitable for the detection of IgG from swine plasma. HPLC method is simple and can be carried out on the qualitative and quantitative detection of IgG .The results have high accuracy and good reprodcuibility.
