脑胶质瘤中CLDN-7基因甲基化状态及蛋白表达的研究 被引量:3

摘要 目的探讨脑胶质瘤血浆中CLDN-7基因的甲基化状况及蛋白表达的关系。方法采用巢式PCR、亚硫酸氢盐测序PCR(BSP)及免疫组织化学技术方法,检测56例脑胶质瘤血清标本中CLDN-7基因启动子区CpG岛的甲基化状态,分析其甲基化程度、蛋白表达和临床资料之间的相关性。结果脑胶质瘤患者血清中CLDN-7基因启动子区甲基化率为35.3%(18/51),按WHO病理分级,其中低级别(Ⅰ~Ⅱ级)组胶质瘤中基因启动子区甲基化率25.0%(9/36)低于高级别(Ⅲ~IV级)组甲基化率60.0%(9/15),两者之间存在显著差别(P<0.05);甲基化蛋白表达阳性率为55.6%(10/18),未甲基化的蛋白阳性表达率为84.8%(28/33)。结论 CLDN-7基因启动子区CpG岛中存在甲基化现象,与脑胶质瘤发生有关;基因的甲基化率与蛋白表达呈负相关关系。 Objective To study the methylation of CLDN-7and protein expression in glioma.Methods nPCR and BSP was adopted to investigate the promoter frequency of these genes in 56 cases of glioma tissue.The CLDN-7 expression in 56 glioma specimens was examined by immunohistochemistry method.The relationship between frequency of methylation of this gene and clinical disease material was analyzed.Results In patientions with glioma,the frequency of methylation in CLDN-7 was 35.3%(18/51).The rate of CLDN-7 promoter methylation in grades Ⅰ~Ⅱ gliomas was significantly lower than that(60.0%)in grades Ⅲ~Ⅳ gliomas(P0.05).Conclusion CLDN-7 promoter(CpG island)hypermethylation exists in glioma,which may correlate with the occurrence of glioma.
