雷公藤多甙对糖尿病大鼠尿蛋白、肾小球硬化及足细胞的影响 被引量:6

Effect of Tripterygium wilfordii polyglucoside on podocytes and urinary protein and glomerulosclerosis in diabetes rats

摘要 目的观察雷公藤多甙(TWP)对糖尿病大鼠尿蛋白、肾小球硬化及足细胞的影响,探讨TWP对糖尿病大鼠肾脏保护作用的效果及可能的机制。方法运用链脲佐菌素(STZ)建立糖尿病大鼠模型,设立TWP组(给予TWP治疗8周)、厄贝沙坦组(给予厄贝沙坦治疗8周)、模型对照组及正常对照组各8只,检测各组的24 h尿蛋白定量、Ⅳ型胶原表达水平以及肾组织中Nephrin蛋白与Podocin蛋白表达水平,并进行比较。结果 8周后各药物干预组24 h尿蛋白定量和肾组织中的Ⅳ型胶原表达水平均低于模型组(P均<0.05),其中TWP组和厄贝沙坦组间的24 h尿蛋白定量比较差异无统计学意义,但TWP组的Ⅳ型胶原表达水平低于厄贝沙坦组(P<0.05);TWP组和厄贝沙坦组Nephrin和Podocin蛋白表达均强于模型对照组,但TWP组和厄贝沙坦组间比较差异无统计学意义。结论 TWP可能通过减少糖尿病大鼠的尿蛋白及减轻其肾小球硬化达到保护肾脏的作用,其机制可能与TWP具有保护肾脏足细胞的功能有关。 Objective To investigate the effect of Tripterygium wilfordii polyglueoside (TWP) on podocytes and glomerulosclerosis in diabetes rats. Methods The diabetes rat model was made by streptozoto-tin (STZ). The rat model was treated with TWP and irbesartan. The rats were randomly divided into the follow-ing groups: the TWP group, irbesartan group, the model group and the normal control group. 24 h urine pro-tein count, the expression of nephfin, podocin and type IV collagen in renal tissue were detected. Results Compared with the normal control group, 24h urine protein count and type IV collagen in kidney tissue were significantly increased in tile model group. The indicators were significantly decreased after drug intervention ( P 〈 0. 05 ). 24h urine protein count was no significant statistical difference in the TWP group and irbesartan group ( P 〉 0. 05 ). Type IV collagen in the TWP group was significantly lower than that in the irbesartan group ( P 〈 0. 05 ). Compared with the normal control group, nephrin and podocin expression in the renal tissue were significantly reduced in the model group. Compared with the model group, nephrin and podocin expression in the TWP group and the irbesartan group were significantly enhanced, but No significant differences between the two groups. Conclusion TWP can alleviate renal tissue damage in diabetes rats by reducing glomerulosclero-sis and urinary protein. The possible mechanism may be that TWP can protect the podocytes in diabetes DN rats.
