肝细胞癌中miR-92a的表达及其对HepG2细胞凋亡的影响 被引量:4

The Expression of miR-92a in Hepatocellular Carcinoma and the Influence of miR-92a on Apoptosis of HepG2 Cell

摘要 目的:探讨miR-92a与肝细胞癌的相关性,及其表达改变对肝细胞癌HepG2细胞凋亡的影响。方法:实时定量PCR检测肝细胞癌和癌旁正常组织中miR-92a的表达;将miR-92a的抑制物miR-92a inhibitor转染了肝细胞癌HepG2细胞,然后检测了转染后细胞的凋亡。结果:与癌旁正常组织相比,肝细胞癌组织中miR-92a的表达显著上调;转染后miR-92a inhibitor组的凋亡率显著增加。结论:miR-92a与肝细胞癌的发生相关,抑制其表达能够促进肝细胞癌细胞凋亡,在肝细胞癌中发挥癌基因的作用。 Objective: To research the relationship of miR-92a expression with hepatocellular carcinoma, and the influence of miR-92a on apoptosis of hepatocellular carcinoma HepG2 cell line. Methods: Real-time PCR was used to detect miR-92a in hepatocellular carcinoma and peri-cancerous tissues, miR-92a inhibitor was transfected to HepG2 cell respectively, and then the apoptosis of HepG2 cell was detected by flow cytometer. Results: The expression of miR-92a in hepatocellular carcinoma was much higher than that in peri-cancerous tissue. After transfection, the apoptosis ratio ofpre-miR-92a group decreased, and the apoptosis ratio of miR-92a inhibitor group increased significantly. Conclusions: miR-92a was related with the development of hepatocellular carcinoma. Inhibiting the expression of miR-92a could accelerate the apoptosis of hepatocellular carcinoma cell. It acted as an oncogene in hepatocellular carcinoma.
