摘要 目的:肿瘤细胞的增殖和发展与多种细胞周期调控因子的异常密切相关,Skp-2通过泛素化降解p27,使肿瘤细胞G1-S期调控发生紊乱,从而参与肿瘤细胞的某些生物学性状,本实验探讨牙龈癌发生发展过程中Skp2蛋白的表达以及其与牙龈癌临床病理学之间的意义。方法:收集同期46例牙龈癌及22例正常牙龈组织标本,制备组织切片,应用SP免疫组化法检测正常黏膜及牙龈癌中Skp2蛋白的表达,以及Skp2蛋白在牙龈癌不同病理分级和临床分期中的表达。结果:Skp2蛋白在正常口腔粘膜组织与牙龈癌中的阳性表达率差异有统计学意义,Skp2蛋白在牙龈癌中的阳性率(32.61%),显著高于正常黏膜组织(4.55%()x2=6.51 P<0.05)。应用阳性表达率表示Skp2与牙龈癌的临床分期、病理分级、性别、年龄之间的关系,结果显示Skp2与牙龈癌的临床分期及病理分级显著相关成正比例关系,差异具有统计学意义(P<0.05),而与年龄、性别等因素不相关,无统计学差异(P>0.05)。结论:Skp2蛋白在口腔牙龈癌的临床分级和病理分级成正比例相关,与年龄、性别无显著相关,Skp2可能在牙龈癌的发生发展中起着重要的正向调控作用,检测Skp2有助于判断牙龈癌的恶性程度及其预后,为临床判断病变程度及治疗提供更加合理的方案。 Objective: Cell cycles of tumor cells' growth and development is closely related to a variety of regulatory factors of the abnormal. This essay is to study the Skp2 protein expression in the development process and the clinical pathological significance on the gingival carcinoma. Methods: 46 cases with gingival cancer and 22 cases with normal gum tissue specimens were collected over the same period to prepare the tissue sections, and the Skp2 protein expression in the normal mucosa and gingival carcinoma with different pathological grades were detected by SP immunohistochemical methods. Results: There were statistical differences between the Skp2 protein expression in normal oral mucosa tissue and gingival carcinoma, Skp2 in gingival cancer-positive rate (32.61%) was siguificantly higher than that in normal mucosa (4.55%) (x^26.51, P〈0.05). The application of positive expression rate of the relationship of Skp2 with gingival cancer clinical stage, histological grade, gender and age. There were significantly associated between the Skp2 protein expression in normal oral mucosa tissue and gingival carcinoma (P〉0.05), but were unrelated with age, gender and other factors (P〉0.05). Conclusion: Skp2 protein in oral gingival cancer clinical grading and pathological grading is proportional to related. No significant correlation with age, gender, Skp2 may occur in the positive regulation effect plays in the development of important gingival carcinoma, detection of Skp2 is helpful to judge the malignant degree and prognosis of carcinoma of gingiva, provide a more reasonable scheme for clinical severity and treatment.
