摘要
采用高效液相色谱法建立了金铁锁药材中尿囊素的含量测定方法。金铁锁药材经乙醇提取后,取续滤液进行分析。采用ZORBAX NH2色谱柱(4.6 mm×150 mm,5μm),检测波长220 nm,柱温40℃,流动相为乙腈-水(93∶7),流速1.0mL.min-1。结果表明,尿囊素线性关系良好,具有较好的精密度和重复性。在0.010 4~0.166 g.L-1,尿囊素的线性相关系数R2=0.999 4(n=5);RSD(n=6)小于2.1%;平均回收率为95.47%~100.9%。该方法具有良好的灵敏度、回收率和重复性,可为金铁锁药材的质量控制提供参考。
A method for determintion of allantion in Psammosilene tunicoides was established by HPLC. Using alcohol as the extraction solvent ,the subsequent filtrate of P. tunicoides was analysed by HPLC. Allantoin was successfully detected and separated by ZORBAX NH2column (4. 6 mm × 150 mm,5 μm)at wavelength of 220 nm and column temperature of 40 ℃, with acetonitrile-water (93: 7) as mobile phase at a flow rate of 1.0 mL. min-1. The results showed that it had a good linear relationship between the con- cent ration of allantion and chromatographic peak area. The linear correlation coefficient of allantion was 0. 999 5 in 0. 010 4-0. 166 g .L-1. The relative standard deviation of six parallel injections was less than 2. 1%. The average recoveries were ranged from 95.47% to 100. 9%. This method was sensitive and accurate for the determination of allantion in P. tunicoides.
