摘要
目的:通过研究四妙勇安汤对动脉粥样硬化(AS)大鼠SOCS1和SOCS3的影响,探讨其抗炎机制。方法:将体质量为200~230 g的纯种SD雄性大白鼠40只按数字表法随机分为假手术组、空白模型组、四妙勇安汤组、普罗布考组,每组10只。制备AS模型,各组于造模后第3天开始给药,四妙勇安汤按15.22 g.kg-1给药,普罗布考按0.16 g.kg-1给药,假手术组、空白模型组给予同体积的生理盐水。于实验16周末禁食16 h后,麻醉动物,腹主动脉取血,分离血清,检测肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)和白介素-10(interleukin-10,IL-10);取大鼠胸腹主动脉段,采用HE染色法观察其病理改变,采用Westen-blot法观察SOCS1和SOCS3蛋白的表达,采用RT-PCR法观察SOCS1和SOCS3mRNA的表达。结果:病理结果提示,本实验成功制备了AS大鼠模型,四妙勇安汤升高IL-10的作用优于普罗布考(P<0.01),而对TNF-α的影响两者则没有显著差异(P>0.05);普罗布考降低SOCS1蛋白的作用优于四妙勇安汤(P<0.01),四妙勇安汤组降低SOCS3蛋白的作用优于普罗布考组(P<0.05);四妙勇安汤和普罗布考均可以降低SOCS1mRNA和SOCS3mRNA的表达,组间无显著差异(P>0.05)。结论:四妙勇安汤的抗炎机制与调节SOCS1和SOCS3表达有关。
OBJECFIVE We want to investigate the mechanism of antiqnflammatory effect by studying the effect of Simiao soup on SOCS1 and SOCS3 in atherosclerosis rats. METHODS 40 purebred male SD rats weighing 200- 230g rats were ran- domly divided into sham operation group, untreated group, Simiao soup group, prohucol group (n = 10). Each group was given with drugs or saline for three days after the AS models were prepared, Simiao soup group was administrated 15. 22 g-kg-1 , probucol group was 0. 16 g.kg-1, sham operated group and untreated group was given the same volume of saline. The serum was separated from the blood which was from abdominal aorta when the animals were anesthetized after 16h fasting at 16th weekend of experiment, and detected the TNF-α and IL-10; HE staining was used to observe the pathological changes, SOCS1 and SOCS3 protein expression was observed by Westerwblot and RT-PCR was used to observe expression of SOCS1 and SOCS3 mRNA in rat aortic. RESULTS The pathological results suggest that as rat model was successfully prepared in this study. The role of Simiao soup group increased IL-10 was superior to probucol group (P〈0. 01), but there was no significant difference to TNF-a (PD0. 1/5); the probucol group reduced SOCS1 protein was greater than Simiao soup group (P〈0. 01), the level of SOCS3 protein in Simiao soup group higher than probueol group (P〈0. 05) ; Simiao soup group and probucol group could reduce the expression of SOCS1 mRNA and SOCS3 mRNA, there was no significant difference (P〉0. 01) between the two groups. CONCLUSION The mechanism of anti-inflammatory effect of Simiao soup may be related to adjust the expression of SOCS1 and SOCS3.
