摘要 目的筛选和鉴定骨肉瘤发病的相关基因,揭示骨肉瘤发病的分子机制。方法收集3例手术切除骨肉瘤新鲜组织标本(用患者正常骨组织作为对照),提取组织总RNA,经逆转录、Cy3及Cy5荧光分子标记和基因芯片杂交,采用经验贝叶斯法筛选出骨肉瘤发病的差异表达基因,最后用RT-PCR和Western blot验证部分基因的表达。结果基因芯片筛选到105个骨肉瘤发病的差异表达基因,其中在骨肉瘤中表达上调的42个,下调的63个,RT-PCR和Western blot验证BAX、HMMR和SHB基因的表达结果与芯片结果完全相符。结论骨肉瘤的发病与BAX、HMMR和SHB等基因的异常表达密切相关。 【Objective】To screen and identify incidence related genes of osteosarcoma.【Methods】The three cases of surgical resection lesion tissue of patients with osteosarcoma were collected,normal bone tissue was used as a comparison.The total RNA was extracted and cDNA was amplified by reverse transcription,the cDNA was labeled by Cy3 and Cy5 in vitro,hybridizing microarray,screening of differentially expressed genes using empirical Bayes methods,finally the expression of the gene was verified by RT-PCR and Western blot.【Results】105 differentially expressed genes,42 up-regulated genes and 63 down-regulated genes in the tumor tissue were microarray screened compared to normal tissue.Gene(BAX;HMMR;SHB) expression by RT-PCR and Western blot exactly matched with the microarray results.【Conclusion】Abnormal expression of the gene BAX,HMMR and SHB is related to the incidence of osteosarcoma closely.
