银杏叶提取物对新生大鼠缺氧缺血性脑损伤后脑内基质金属蛋白酶的影响

Effect of Ginkgo biloba extract on the expression of matrix metalloproteinase in the hypoxic ischemic brain damage

摘要 目的:观察银杏叶提取物(EGb)对新生大鼠缺氧缺血性脑损伤(HIBD)后脑内基质金属蛋白酶表达的影响。方法:7日龄SD幼鼠32只随机分为正常对照组(ZC组)、假手术组(SH组)、银杏叶提取物干预组(EGb组)、生理盐水对照组(NS组),采用经典的Rice法制作HIBD模型,ZC组不做任何处理,EGb组模型建立后即可腹腔注射银杏内酯B 10 mg/kg,每日1次,连续注射10天,NS组每日腹腔注射生理盐水共10天,采用免疫组化方法观察大脑皮质MMP-2、MMP-9的分布及表达。结果:NS组与ZC组相比,MMP-2、MMP-9在大鼠脑皮质层表达明显增加(P<0.01),MMP-2在第4天表达最为明显,MMP-9在24 h时表达最为明显;EGb组与NS组各个时间段相比,MMP-2、MMP-9表达明显减少(P<0.01)。结论:EGb可减少新生大鼠缺氧缺血性脑损伤后的MMP-2、MMP-9的表达,对缺氧缺血性脑损伤新生大鼠脑有神经保护作用。 Objective:To study the effect of extracts of Ginkgo biloba leaf(EGb) on expression of matrix metalloproteinases(MMPs) in neonatal rat brain after hypoxic-ischemic brain damage(HIBD).Methods:Seven-day-old SD rats were randomly divided into normal control(ZC),sham(SH),HIBD(NS),EGb(EGb) groups.HIBD model was induced by the classical Rice method.Rats in ZC group were given without any treatment.Rats in EGb group received intraperitoneal injection of ginkgolide B 10 mg/kg.Rats in NS group received an intraperitoneal injection of saline,once daily,for ten consecutive days.The distribution and expressions of MMP-2 and MMP-9 in cerebral cortex were observed by immunochemical method.Results:Compared with ZC group,expressions of MMP-2,MMP-9 in rat cortical cortex were significantly increased in NS group(P&lt;0.05).Expression of MMP-2 was most obvious in the fourth days,expression of MMP-9 was most obvious on 24 hours after made model.Compared with NS group,expressions of MMP-2,MMP-9 in EGb group at each time period were significantly reduced(P&lt;0.01).Conclusion:EGb can reduce expressions of MMP-2 and MMP-9 in neonatal rat brain cortex after HIBD,and has protective effect on HIBD.
