摘要
目的建立HPLC-ELSD法测定乌灵菌粉中甘露醇的含量。方法采用氨基柱(Ultimate XB-NH2,4.6mm×250mm,5μm),柱温:25℃,进样量:20μL,以乙腈-水(84∶16)为流动相洗脱,流速:1.0mL.min-1;ELSD漂移管温度:95℃,氮气压力:30psi。结果甘露醇在332～1992μg.mL-1浓度范围内线性关系良好(r=0.9992,n=6);方法回收率为98.86%,RSD为1.01%。结论该方法准确度高、专属性强、重复性好,可用于乌灵菌粉的质量控制。
Objective To establish an HPLC-ELSD method to determine the content of mannitol in Wuling Powder. Methods The determination was performed on an amino chromatographic column （Ultimate XB-NH2, 4.6 min×250 mm, 5μm） with a mobile phase of acetonitrile-water （84 ： 16）. The flow rate was 1.0 mL · min^-1, the temperature of the ELSD drift tube was set at 95℃, and the nitrogen pressure was 30 psi. The injection dosage was 20 μL. Results A good linear correlation （r = 0. 9992） was observed when the content of mannitol was in the range of 332-1992 μg ·mL^-1. The recovery of the method was 98.86%, and the relative standard deviation （RSD） was 1.01% （n=6）. Conclusion This method is reliable, accurate, with high sensitivity and repeatability. It can be used for quality control of Wuling Powder.
