HPLC法同时测定耳聋胶囊中龙胆苦苷和黄芩苷的含量

Simultaneous Determination of Gentiopicrin and Baicalin in Erlong Capsules by HPLC

目的建立HPLC法同时测定耳聋胶囊中龙胆苦苷和黄芩苷的含量。方法采用Dikma ODS C18(迪马公司;4.6mm×200mm,5μm)色谱柱;甲醇-0.2%磷酸梯度洗脱,流速:1mL·min-1;检测波长:270nm;柱温:30℃。结果龙胆苦苷和黄芩苷分别在0.01165～0.1745μg(r=0.9999,n=5)和0.10072～1.5108μg(r=0.9999,n=5)范围内有良好的线性关系,平均加样回收率分别为97.5%和98.2%(n=6)。结论本方法简单、快速、准确,可用于耳聋胶囊的质量控制。

Objective To establish an HPLC method to simultaneously determine the contents of gentiopicrin and baicalin in Erlong capsules. Methods The analysis was performed on a Dikma ODS C18 column （Dikma Inc.; 200 min×200mm,5μm） with gradient elution of methanol and 0.2% phosphoric acid at a flow rate of 1.0 mL ·min-1. The UV detection wavelength was 270 nm and the column temperature was set at 30 ℃. Results The linear ranges were 0. 01165-0. 1745μg （r=0. 9999, n=5） for gentiopicrin and 0. 10072-1. 5108μg （r = 0. 9999, n = 5） for baicalin. The average recoveries of gentiopicrin and baicalin were 97.5% and 98. 2% （n =6）, respectively. Conclusion The method is simple, rapid and accurate, which is suitable for quality control of Erlong capsules.
