摘要
目的探讨吡格列酮对大鼠脑出血后脑组织水通道蛋白4(AQP4)表达的影响。方法将40只成年雄性Wistar大鼠随机分为假手术组、脑出血组、吡格列酮组。应用自体血尾状核定位注射方法建立大鼠自体血脑出血模型,分别采用免疫组化及半定量逆转录聚合酶链反应方法,检测脑组织AQP4蛋白及mRNA的表达。结果脑出血组AQP4蛋白的表达较假手术组明显升高(F=222.03,q=29.15,P<0.05),而吡格列酮组AQP4蛋白的表达较脑出血组明显降低(q=9.21,P<0.05)。脑出血组AQP4mRNA含量较假手术组明显增高(F=327.22,q=33.13,P<0.05);吡格列酮组AQP4mRNA含量与脑出血组比较明显降低(q=3.98,P<0.05)。结论吡格列酮能够抑制脑出血后血肿周围脑组织AQP4的表达,从而减轻脑水肿。
Objective To study the effects of pioglitazone on aquaporin-4 (AQP4) in rats after intracerebral hemorrhage. Methods Fourty adult male Wistar rats were randomly divided into sham-operated group, intracerebral hemorrhage model group ( model group) and pioglitazone group. Models of cerebral hemorrhage were created. Immunohistochemistry and RT PCR were used to detect the expressions of AQP4 and mRNA in the brain tissues. Results The expression of AQP4 protein in the model group was higher than that in the sham operated group (F=222.03,q=29.15,P〈0.05), and that in the pioglitazone group was lower than that in the model group (q=9.21 ,P〈0.05). Compared with the model group, the level of AQP4 mRNA in the sham-operated group (F=327.22,q=33.13,P〈0.05), and that in pioglitazone group were lower (q=3.98,P〈0.05). Conclusion Pioglita zone can inhibit the expression of AQP4 in brain tissue surrounding hematoma after cerebral hemorrhage, and suppress the expres- sion of AQP4 resulting in relief of brain edema.
