壮骨颗粒对骨折愈合中HIF-1α分泌及骨痂形成影响 被引量:1

THE EFFECT OF ZHUANGGU GRANULA ON SECRETION OF HIF-1α AND CALLUS FORMATION IN FRACTURE HEALING

摘要 目的探讨壮骨颗粒对动物模型骨折断端缺氧诱导因子-1α(HIF-1α)分泌与骨痂形成的影响。方法32只新西兰大白兔随机分成壮骨颗粒组和对照组,各16只。建立兔左前肢桡骨骨折模型,壮骨颗粒组给予壮骨颗粒灌胃治疗,对照组给予安慰剂,分别于术后1、2、4、8周拍X线片,盲法对外骨痂和骨折线进行定量评价;取骨折处骨标本,行免疫组化染色,检测骨痂组织HIF-1α阳性强度。结果壮骨颗粒组、对照组外骨痂与骨折线评分随时间延长逐渐增加,差异有显著性(F=555.63、458.82,P<0.01);从第2周开始,壮骨颗粒组与对照组外骨痂与骨折线评分比较,差异有显著性(F=6.51~134.14,P<0.05)。骨折后第1周两组均未见HIF-1α阳性染色;骨折2周后,两组组内不同时间比较、两组间同一时间HIF-1α阳性强度比较,差异均有显著性(F=6.76~64.93,P<0.05)。结论壮骨颗粒可促进HIF-1α在骨痂组织中的表达,加速骨痂生成,提高骨痂质量。 Objective To investigate the effect of Zhuanggu granula on secretion of HIF let and callus formation in animal model with fracture. Methods A total of 32 New Zealand rabbits were equally randomized to two groups. A model of left radius fracture was created in rabbits. Intragastric administration of Zhunggu granula was given to rabbits in the Zhunggu granula group, and to those in the control, placebo was offered. X-ray films were taken 1, 2, 4 and 8 weeks after surgery. The blind method was used to quantitatively assess external callus and fracture line. Bone samples from the fractured part were taken and stained by immunohistochemisty to determine positive HIF-1α in bony callus. Results The score of external callus and fracture line in both groups was increased along with the extension of duration, the different was significant. The differences of scores of external callus and fracture line between Zhuanggu granula and control group were significant beginning from the second week (F=6.51- 134.14, P〈0.05). No positive staining of HIF-1α was found on the first week of fracture; after two weeks, the positive expression of HIF-1α of different time points in the same group, and that of the same time point between the two groups were significant (F= 6.76-64.93,P〈0.05). Conclusion Zhuanggu granula can promote the secretion of HIF-1α in bone callus, and accelerate its formation and raise its quality.
