腺瘤性大肠息肉中DNA甲基转移酶mRNA的表达改变

摘要 目的研究三种DNA甲基转移酶(DNA methyltranferase,DNMT)在腺瘤性大肠息肉发生发展中的作用。方法从7例腺瘤性大肠息肉和7例正常大肠粘膜组织中提取RNA后反转录成cDNA,beta-actin作为内参,利用real-time PCR技术对三种DNMT的表达情况进行。结果腺瘤性大肠息肉和正常粘膜组织中DNMT1 mRNA对β-actin mRNA的相对丰度值分别是1.02×10-3±4.87×10-4和8.67×10-4±2.18×10-4,大肠息肉组织中与正常粘膜组织中DNMT1表达无有显著性差异(P>0.05);腺瘤性大肠息肉和正常粘膜组织中DNMT3A mRNA对β-actin mRNA的相对丰度值分别是3.26×10-2±4.96×10-3和3.00×10-2±5.20×10-4,它们之间无显著性差异(P>0.05);腺瘤性大肠息肉和正常粘膜组织中DNMT3B mRNA对β-actin mRNA的相对丰度值分别是2.95×10-1±0.60×10-2和8.78×10-2±0.13×10-2,大肠息肉组织中与正常粘膜组织中DNMT3B表达有显著差异(P<0.05)。结论 DNMT3B可能在腺瘤性大肠息肉的发生中起到了重要作用。 Objective: To study the roles of three of kinds of DNA methyltransferases (DNMTs) in the occurring of colorectal adenomatous polyps. Methods: Three kinds of DNMTs mRNA level were detected by real - time PCR in 7 cases of eolorectal adenomatous polyps (CAP) and 7 cases of normal mucosal tissue (N). Results : The relative abundance of DNMT1 mRNA to BETA - ACTIN mRNA in colorectal adenomatous polyps and in normal mucosal tissue were 1.02 ×10 -3 ±4.87× 10 -4 and 8.67 × 10 -4 ±2.18 × 10 -4 respectively. The relative abundance of DNMT3A mRNA to BETA - ACTIN mRNA in colorectal adenomatous polyps and in normal mucosal tissue were3.26 × 10-2 +4.96 × 10-3 and 3.00 × 10-2 ±5.20 × 10-4, respectively. The DNMT1 and DNMT3A mRNA were not found to be increased in coloreetal adenomatous polyps ( P 〉 0.05 ,vs normal). The relative abundance of DNMT3B mRNA to BETA - ACTIN mRNA in colorectal adenomatous polyps and in normal mucosal tissue were 2.95 × 10 -1 + 0.60× 10-2 and 8.78 ×10-2 ±0.13 ×10 -2 respectively. The DNMT3B mRNA was found to be increased in colorectal adenomatous polyps (P 〈 0.05, vs normal mucosal tissue). Conclusion: The increasing expression of DNMT3B may play an important role in the occurring of colorectal adenomatous polyps.
