骨桥蛋白和CD44v6在口腔鳞状细胞癌中的表达及其意义

EXPRESSION AND SIGNIFICANCE OF OSTEOPONTIN AND CD44v6 IN ORAL SQUAMOUS CELL CARCINOMA

目的探讨骨桥蛋白(osteopontin,OPN)和黏附分子CD44拼构变异体6(homing cell adhesion moleculevariant 6,CD44v6)在口腔鳞状细胞癌中的表达及其临床意义。方法采用免疫组织化学链霉素抗生物素蛋白-过氧化物酶连结(SP)法检测OPN和CD44v6蛋白在口腔鳞状细胞癌组织中的表达,分析其表达与患者临床病理学资料之间的关系。结果 OPN和CD44v6在口腔鳞状细胞癌中阳性表达率分别为64.3%、61.2%,均明显高于在癌旁正常口腔黏膜中的表达(P<0.01)。OPN和CD44v6组织分化Ⅲ级的阳性率明显高于Ⅰ～Ⅱ级,淋巴结转移组阳性率明显高于淋巴结未转移组,差异有统计学意义(P<0.05),但与性别、年龄无关。结论 OPN和CD44v6与口腔鳞状细胞癌的分化程度和淋巴结转移关系密切,在其进展过程中起着重要的作用;而OPN与CD44v6的关系有待进一步研究。

Objective To investigate the expression of osteopontin （OPN） and CD44v6 and their relationship with clinical pathological characteristics in oral squamous cell carcinoma （OSCC）. Methods The expression of OPN and CD44v6 proteins in oral squamous cell carcinoma were detected by SP immunohistochemical method. According to the clinical information, all data were analyzed by SPSS13.0 for windows to explore the relationship between the two indexes and the pathological features. Results The positive rates of OPN and CD44v6 in oral squamous cell carcinoma were respectively 64.3% and 61.2% , which were significantly higher compared with those of adjacent non-tumor tissues and normal oral membrane tissues （P 〈 0.01 ）. The positive rate expression of OPN and CD44v6 of poor differentiation group were significantly higher than those of well differentiation group and moderation differentiation group （ P 〈 0. 05 ）. The metastasis of lymph node, the expression levels of OPN and CI）44v6 in lymph node metastasis group were significantly higher than those of non lymph node metastasis group （ P 〈 0.05 ） , however, no significant difference existed in gender and age. Conclusion The expression of OPN and CD44v6 are closely related metastasis of oral squamous cell carcinoma, and they squamous cell carcinoma, and the relationship between with the differentiation degree, lymphatic node play important roles in the development of oral OPN and CD44v6 gen needs further studies.
