缺血/再灌注损伤心肌Intermedin及其受体mRNA表达水平的动态变化

摘要 目的利用大鼠在体心肌缺血/再灌注(I/R)损伤模型,观察再灌注过程中缺血区和非缺血区心肌Intermedin(IMD)及其受体mRNA的表达水平。方法结扎左前降支冠状动脉制备大鼠心肌在体I/R损伤模型,检测血流动力学指标和血浆LDH活性,RT-PCR法检测IMD及其受体mRNA表达水平。结果与对照组相比,I/R组大鼠心功能显著降低,血浆乳酸脱氢酶活性显著升高。缺血区心肌CRLR和RAMP2的mRNA表达水平分别在再灌注12小时和24小时显著下调,RMAP1和RAMP3的mRNA表达水平分别在再灌注6小时和24小时显著上调;非缺血区心肌CRLR和RMAP3的mRNA表达水平显著上调,RAMP1和RAMP2的mRNA表达水平无显著变化。I/R组大鼠心肌IMD的mRNA表达水平在缺血区和非缺血区均显著上调。结论 IMD及其受体系统是重要的内源性心血管保护系统,在拮抗心肌I/R损伤中发挥重要作用。
