采用嗜热吸水链霉菌单一抗原ST-omlA诱导小鼠大棚肺模型 被引量:2

摘要 目的探讨纯化的嗜热吸水链霉菌单一表面抗原能否诱导C57BL/6小鼠大棚肺模型。方法 C57BL/6小鼠20只,8周龄,雌性,体质量25~30 g,完全随机分为模型组和对照组,每组10只。模型组:纯化的嗜热吸水链霉菌表面抗原ST-omlA 0.2 mL(1 mg/mL)鼻内滴注,连续3 d/周,连续滴注4周;对照组:0.9%无菌生理盐水0.2 mL鼻内滴注,连续3 d/周,连续滴注4周。结果①组织病理学改变:模型组第2周末,可见肺泡炎改变;第3周末,可见肉芽肿改变;第4周末,可见纤维化改变。对照组无异常。②免疫组织化学染色结果:模型组ST-omlA滴注后的第4周末,Ⅰ型胶原蛋白及血管内皮生长因子表达明显增多,呈阳性;对照组Ⅰ型胶原蛋白及血管内皮生长因子表达呈阴性。结论纯化的嗜热吸水链霉菌表面抗原ST-omlA抗原制剂可以诱导C57BL/6小鼠大棚肺模型。 【Objective】 To explore whether ST-omlA-the purified single antigen of the streptomyces thermohydroscopicus can induce the green-house lung.【Methods】 Twenty C57BL/6 mice(female,weighted 25 to 30 g) were randomly divided into two groups(10 mice per group): Group model:intranasal instilled ST-omlA 0.2 mL(1 mg/mL) during 3 consecutive days per week,4 consecutive weeks;Group control: intranasal instilled 0.2 mL 0.9% saline during 3 consecutive days per week,4 consecutive weeks.【Results】 Pathology change: Group model: the 2nd week,it appeared alveolitis;the 3rd week,granuloma consist edl;the 4th week,the number of fibroblasts increased.Group control: it’s normal during the 4 weeks.IHC: Group model: Col Ⅰ and VEGF was positive at the end of intranasal instilled ST-omlA;Group control: Col Ⅰ and VEGF was negative at the end of intranasal instilled 0.9% saline.【Conclusion】 Intranasal instilled of ST-omlA,the purified single antigen of the streptomyces thermohydroscopicus,can induce the green-house lung.
