摘要
为研究用碱法提取蚂蚁蛋白及用单酶水解的方法提取蚂蚁蛋白,考察其最佳工艺条件。采用L9(33)正交试验设计方法提取蚂蚁蛋白,分别对碱液浓度、浸泡温度、固液比进行实验与结果分析,以确定最佳溶解条件为:碱液浓度1.5%,浸泡温度80℃,固液比1:20,蚂蚁蛋白提取率达18.82%。然后采用L9(34)蚂蚁蛋白酶水解正交试验设计方法,对酶种类、温度、时间、固液比行实验与结果分析,确定胰蛋白酶为较适宜酶,最佳水解条件为:pH8.0,酶量2.0%,反应温度40℃,固液比1:7.5,反应时间5h,水解率达19.28%。用单酶水解的方法提取蚂蚁蛋白提取率高。
To investigate the optimal synthesizing condition of the ant protein extraction by alkali process and single enzyme reaction. The alkali process extraction designed by L9(33) orthogonal method including measurement and analysis on lye concentration, soaking temperature (℃) and solid-liquid ratio. The ant protein extraction reached to 18.82%under the best dissolution condition of lye concentration 1.5%, soaking temperature 80℃, solid-liquid ratio 1:20. While In the single enzyme reaction, the kind of enzyme, temperature, time and solid-liquid ratio of the ant protein protease hydrolysis were tested and analyzed by L9(34) orthogonal method. Trypsin were obtained to be the comparatively suitable enzyme, and its hydrolysis rate reached to 19.28% under the optimal hydrolysis condition of pH 8.0, enzyme concentration 2.0%, temperature 40℃, solid-liquid ratio 1:7.5, total hydrolysis time 4 h. Conclusion: Single enzyme reaction improving the extraction rate of ant protein.
