中间香型烤烟叶面腺毛形态及分泌特性研究 被引量:15

摘要 以贵州省遵义市中间香型烟叶典型生态区的代表品种K326为材料,采用显微观察、GC/MS及RT-PCR技术,对叶面腺毛进行了形态观察、化学成分检测及基因表达分析。结果表明:腺毛密度随叶片发育呈下降趋势,在叶龄20 d时下降幅度最大;腺毛主要分泌物中西柏烷类化合物含量最高,并在叶龄70 d达到最高水平;西柏烷类化合物合成关键基因西柏三烯一醇合成酶基因(cembratriene-ol synthase gene,CYC-1)及西柏三烯一醇羟化酶基因(cembratriene-ol hydroxylase gene,CYP71D16)在20 d、40 d和70 d时表达较强,呈现出与化学成分变化类似的趋势,推测认为CYC-1和CYP71D16的表达可影响西柏烷类化合物的含量。 Microscopic observation, GC/MS, and RT-PCR techniques were employed to study morphology, surface chemistry feature and relevant gene expression level of K326 tobacco leaf trichome from Zunyi, Guizhou province, where neutral flavor tobacco leaf was grown. Results showed that trichome density gradually decreased during growing stage and showed the sharpest decline at 20d leaf age. Cembratriendiol group was the most abundant component in leaf trichome exudates, and its content reached a peak at 70d leaf age. The relevant genes, i.e. cembratriene-ol synthase gene (CYC- 1) and cembratriene-ol hydroxylase gene (CYP71D 16), showed similar expression tendency, which suggested that they can affect content of cembratriendiol group.
