摘要
以L-谷氨酰胺为手性选择剂,建立了对普萘洛尔、慢心律、心律平、苯丙哌酮、异丙肾上腺素、氯胺酮、山莨菪碱、洛贝林、布比卡因、苯海索等10种手性药物对映体的毛细管电泳快速分离方法,在含有60mmol/L L-谷氨酰胺,60mmol/L硼砂(pH=9.25)的缓冲溶液中,10种手性药物在11min内达到基线分离。并对分离机理作了初步探讨。
A rapid separation method using L-glutamine as chiral selector was set up. Ten kinds of racemates, such as propranolol, mexileine, propafeone,eprajinome, isoprenaline,ketamine, anisodamine,lobeline, bupivacaine, benzhexol ect. were successfully separated by choosing the best separating conditions. The use of borax and L-glutamine as buffer additive was found to be effective for the chiral separation. The pH of the background electrolyte and the concentrations of chiral selector and borax were optimized. In a solution containing 60 mmol/L L-glutamine and 60 mmol/L borax (pH = 9.25), the ten drugs were baseline separated in less than 11 minutes. The separating mechanism was also discussed.
