摘要
目的检测bcl -2和bax基因蛋白在乳癌中的表达 ,以探讨其在乳癌发生、发展中的作用及其临床病理学意义。方法40例乳癌和 3 1例乳腺良性病变 ,用免疫组化S -P法检测其bcl-2和bax的表达。结果bcl-2基因蛋白在乳癌中心、癌旁、良性病变表达阳性率分别为 72 .5 %( 2 9/ 40 ) ,45 .0 ( 18/ 40 )和 96.8%( 3 0 / 3 1) ( P <0 .0 1) ;按肿瘤细胞分级Ⅰ、Ⅱ、Ⅲ级的bcl -2表达强度分别为 ( 2 .0 8± 0 .2 9) ,( 1.2 3± 0 .2 8)和( 0 .73± 0 .2 3 ) (P <0 .0 1)。bax基因在乳癌中心、癌旁、良性病变中的表达强度分别为 ( 0 .88±0 .13 ) ,( 1.2 0± 0 .10 )和 ( 1.16± 0 .17) (P <0 .0 5 )。结论bcl-2基因蛋白在乳癌中高表达 ,bax基因蛋白则为低表达。说明其细胞凋亡受抑制。bcl-2的高表达可部分反映乳癌的生物学行为和预后良好。
Objective To explore the clinico-pathological significance of bcl-2 and bax oncoprotein exression in the growth and development of breast carcinoma.Methods The expressions of bcl-2 and bax oncoproteins in the specimen from forty cases of primary breast cancers and para-carcinoma as well as thirty-one cases of benign breast lesions were detected by immunohistochemical S-P method.Results The positive expressions of bcl-2 oncoprotein in the primary breast cancers,para-carcinoma tissues and benign lesion tissues were 72.5%(29/40),45.0%(18/40) and 96.8(30/31) respectively;whereas the scores of staining intensity of bcl-2 in the tumors of Grade Ⅰ,Grade Ⅱ and Grade Ⅲ were (2.08±0.29),(1.23±0.28) and (0.73±0.23) respectively(P< 0.01 ).The expression scores of staining intensity from bax gene in the primary breast cancers,para-carcinoma and benign lesion tissues were (0.88±0.13),(1.20±0.10) and (1.16±0.17) correspondingly(P<0.05).Conclusions As compared to the para-carcinoma tissues,the expression of bcl-2 oncoprotein is increased in the breast carcinoma tissues,while that of bax oncoprotein is decreased.It may imply that bcl-2 and bax oncoprotein play an important role in the growth and development of breast carcinoma.High level of Bcl-2 oncoprotein expression may partly reflect the biological behavior of breast cancer and a favorable prognosis.
