猪苓多糖硫酸酯的制备及活性测定 被引量:3

摘要 目的:制备猪苓多糖硫酸酯,同时测定其活性。方法:采用醇沉分级、大孔吸附树脂、聚酰胺树脂和脱蛋白法从中药猪苓中提取分离得到猪苓多糖,硫磺酸-吡啶法进行硫酸酯化,经Sephadex G-75、透析得到硫酸化猪苓多糖,并体外观察硫酸酯化多糖对羟基自由基清除作用。结果:猪苓多糖硫酸酯化前后均有清除Fenton反应由Fe2+-H2O2体系产生的羟自由基,硫酸酯化猪苓多糖对羟自由基的清除能力增强,并呈明显量效关系。结论:制备猪苓多糖硫酸酯的方法准确可靠,为猪苓多糖的进一步开发利用提供实验依据。 Objective: To prepare sulfated polysaccharides from Polyporus umbellatus and investigate its anti-oxidation activity.Method: Polysaccharides was extracted and isolated by alcohol precipitation,macroporous resin,polyamide resin,deproteinization from P.umbellatus.Polysaccharides was sulfated by sulfuric acidpyridine method,sulfated polysaccharides was depurated by Sephadex G-75 and dialysis,and in vitro scavenging effect of sulfated polysaccharides for hydroxyl free radical was observed.Result: Before and after sulfated,polysaccharides from P.umbellatus showed good antioxidation in scavenging hydroxyl free radical from Fe2 +-H2O2 system,scavenging capacity of sulfated polysaccharides for hydroxyl free radical enhanced,and showed a clear dose-effect relationship.Conclusion: This method of preparing sulfated polysaccharides was accurate and reliable,it could provide references for further development and utilization of polysaccharides from P.umbellatus.
